摘  要:用从生产手性药物工厂附近的土壤中筛选出的17株细菌,对布洛芬氯乙酯拆分进行了初步研究,实验结果显示,在LB培养基中,17株细菌分别在12~36h达到对数生长期;它们都有不同程度的水解手性化合物的能力,其中有的还具有较强的立体选择性;17株细菌中只有7株的脂肪酶活性较高,但脂肪酶活性与转化能力及立体选择性不一致.Microorganisms were screened for the production of chiral medicinal product. The bacteria were isolated from soil samples in the vicinity of the factory producing the chiral drug. We obtained 17 strains in the solid and liquid medium containing Tween 80. The activity of lipase was determined and the ratio of ( S+,R- )profen was tested. Experimental results show that these bacteria have different logarithmic growing phase which range from 12 to 36 hours. When the esterified substrate was used, ( S+,R- )profen enantioselective products were produced. Some strains of the bacteria can produce R - profen, while others can produce S + profen. Among the 17 screened strains of bacteria we found 7 strains of high lipase activity and the range of lipase activity is from 136 to 800 μmol NPP/mL at 5min. The lipase is found to be extracellular.
