摘 要:目的通过对人上皮性卵巢癌细胞株3AO在紫杉醇及紫杉醇联合2-(4-吗琳基)-8-苯基-4氢-1-苯并毗喃-4-酮(LY294002)作用后细胞周期蛋白D1(CyclinD1)的表达及细胞周期变化和细胞凋亡的分析,为卵巢癌化疗寻求一种新的高效辅助治疗手段及一种高效的生物学标志物。方法采用RT-PCR技术,检测3AO在空白组、单纯紫杉醇组、和LY294002预处理24h后紫杉醇(紫杉醇+LY294002)作用组的CyclinD1的表达。并采用流式细胞术分析3组CyclinD1的表达水平及细胞凋亡及细胞周期的变化。结果 3AO在空白组、紫杉醇组和紫杉醇联合LY294002组CyclinD1mRNA表达水平不同,呈逐渐减弱趋势。空白组、紫杉醇组和紫杉醇联合LY294002组细胞周期各时相、细胞凋亡率及细胞增殖指数PI(S+G2/M)值不同,细胞凋亡率逐渐增加,细胞增殖指数PI值逐渐下降,细胞生长受到抑制,CyclinD1表达水平逐渐下降。结论卵巢癌3AO中存在CyclinD1基因的异常扩增及过表达,CyclinD1的表达水平与细胞增殖呈正相关。紫杉醇联合LY294002能促进3AO凋亡,LY294002对紫杉醇诱导3AO凋亡具有协同效果。Objective To analyse the expression of CyclinD1 and the change of cell cycle and cell apoptosis in human epithelial ovarian carcinoma cell strain 3AO after treated with Paclitaxe and combined therapy of Paclitaxel and LY29400,thus to explore an efficient treatment method for ovarian cancer.Methods 3AO were treated with Paclitaxel or Paclitaxel combined with LY29400,culture medium was used as blank control.Reverse transcription polymerase chain reaction(RT-PCR)method and FCM were used to explore the expression of CyclinD1,the change of cell cycle and cell apoptosis in the three groups.Results The level of cyclinD1 was lowest in Paclitaxel combine with LY29400 group and highest in blank control group,the changes of PI(S+G2/M)level was the same with cyclinD1.The rate of cell apoptosis was highest in the combined treatment of Paclitaxel and LY29400 and lowest in blank control,and the cell cycle had the same change.Conclusions The level of cyclinD1 was over expressed in 3AO,cyclinD1 expression was positively correlated with cell proliferation,the combined treatment of Paclitaxel and LY29400 may contribute to cell apoptosis of LY29400 may contribute to the Paclitaxel-induced apoptosis.
