摘 要:目的该研究构建重组基因疫苗pcDNA-MAGE-1和pcDNA-hIL-18;探讨联合应用基因疫苗体外抗肝癌免疫治疗的作用。方法基因疫苗分组免疫实验小鼠,在体外验证疫苗免疫后的小鼠脾细胞对MAGE-1阳性和阴性肝癌细胞株的杀伤作用;分别以不同剂量的pcDNA-hIL-18与pcDNA-MAGE-1组合测定免疫鼠脾细胞杀伤率,验证产生高杀伤效应的最佳的pcDNA-hIL-18浓度。结果重组基因疫苗免疫组小鼠脾细胞对两种MAGE-1阳性肿瘤细胞(SMMC-7721、BEL-7402)均有明显的杀伤作用,其杀伤效应与生理盐水组(空白对照),空质粒pcDNA3组(阴性对照)相比差异具有显著性(P<0.01);重组基因疫苗免疫后的小鼠脾细胞对MAGE-1阴性肿瘤细胞(QQY-7701)有一定的杀伤作用,与空白对照及阴性对照免疫组相比差异有显著性(P<0.05);pcDNA-hIL-18+pcDNA-MAGE-1组以(50+100)μg剂量免疫小鼠的脾细胞杀伤效应最高,与pcDNA-MAGE-1和pcDNA-hIL-18组相比差异有显著性(P<0.01)。结论联合应用MAGE-1与IL-18基因疫苗对MAGE-1阳性肿瘤细胞有明显的杀伤作用。【Objective】 To construct recombination gene vaccine pc-MAGE-1 and pc-hIL-18 and to probe anti-cancer effect of the liver of the combined genetic vaccine in vitro.【Methods】 Genetic vaccines were immunized to mice,whose spleen cells were seen as effector cell,and SMMC-7721,BEL-7402,QQY-7701 hepatoma carcinoma cells as target cell,then lethal effect of spleen cell to MAGE(+) and MAGE(-) tumor cells were validated.Kill ratio of different dose pcDNA-hIL-18 + pcDNA-MAGE-1 was detected for the best immune dose.【Results】 Genetic vaccine groups had an apparente lethal effect on MAGE(+) tumor cells,and they had significant deviation from control groups(P 0.01).Kill ratio obviously heightened(P 0.05),lethal effect of pcDNA-hIL-18 + pcDNA-MAGE-1 group was obviously higher than other groups,it was statistically significantly different to pcDNA-MAGE-1 group(P 0.05).Meanwhile,genetic vaccine groups had determinate lethal effect on MAGE(-) tumor cells,and they had a statistically significant difference from control groups(P 0.05).Kill ratio of pcDNA-hIL-18 + pcDNA-MAGE-1(50+100)μg group was highest and was statistically significantly different to pcDNA-MAGE-1 and other groups(P 0.01).【Conclusions】 Lethal effect of pcDNA-hIL-18 + pcDNA-MAGE-1 group is highest on MAGE(+) tumor cells,and combination of IL-18 and MAGE-1 can further reinforce immuno-competence of vaccine.
