摘 要:以欧洲山梅花(Philadelphus coronaries Aureus)、庆典山梅花(Philadelphus‘Galahad’)、东北山梅花(P.schrenkii Rupr.)、京山梅花(P.pekinensis Rupr.)4个山梅花植物为试材,采用正交试验方法对影响花粉生活力的3个因素(蔗糖、硼酸和培养温度)进行优化试验。结果表明:4种山梅花的花粉萌发率在种间存在差异。欧洲山梅花和庆典山梅花花粉萌发率低于京山梅花和东北山梅花。3个因素对东北山梅花花粉萌发率的影响,从大到小的顺序为硼酸、蔗糖、培养温;3个因素对其余3种山梅花花粉萌发率的影响,从大到小的顺序为蔗糖、硼酸、培养温度;培养温度在20~30℃范围内,对花粉萌发率影响不显著。依据正交试验不同组合的花粉萌发率,确定4种山梅花的最佳花粉萌发条件。在此基础上,在3种不同贮藏温度下(常温(20℃)、冷藏(3℃)和冷冻(-23℃)),对花粉生命力进行单因素对比试验,结果表明:3℃低温冷藏为4种山梅花花粉贮藏的最佳贮藏温度。Philadelphus coronaries Aureus, Philadelphus ' Galahad' , P. schrenkii Rupr. and P. pekinensis Rupr. were used as experimental materials to study the effects of culture media ( sucrose and boric acid) and storage temperature on pollen viability. Orthogonal design of three factors at three levels (sucrose content, boric acid content and culture temperature) was applied to optimize the conditions of pollen germination. Results show that there are differences in pollen germination rate between different species. The germination rates of P. coronaries and P. ' Galahad' were lower than those of P. schrenkii. and P. pekinensis. Boric acid content showed the most obvious effect on pollen germination rate of P. schrenkii, followed by sucrose content and culture temperature. However, sucrose exhibited the most obvious effect on pollen germination rate of the other three species, followed by boric acid and culture temperature. The culture temperature ranging from 20 to 30 degrees C bad no significant effect on pellen germination rate. The suitable conditions for pollen germination of the four species were obtained according to the results of orthogonal design. A single factor test was used to optimize the storage conditions of pollen germination at three storage temperatures ( -23, 3, and 20 degrees C). The result shows that the best storage temperature for the four species of Philadelphus is 3 degrees C.
