摘  要:目的观察雷公藤红素对人类急性髓细胞白血病(acute myeloid leukemia,AML)细胞增殖和凋亡的影响。方法初诊人AML细胞按雷公藤红素浓度分成5组,A组为阴性对照组,B~E组分别为雷公藤红素1,2,5,10μmol/L处理组,应用四甲基偶氮唑兰比色法及流式细胞术等方法检测雷公藤红素对细胞的生长增殖抑制作用及诱导凋亡作用。结果作用不同时间后,B~E组增殖抑制率随雷公藤红素浓度的增加而增加,与A组比较差异均有统计学意义(P<0.01);C,D组增殖抑制率与B组比较差异有统计学意义(P<0.01),与E组比较差异无统计学意义(P>0.05);雷公藤红素抑制作用的最适浓度为2μmol/L;C~E组作用6,12,24h时增殖抑制率比较差异无统计学意义(P>0.05),与3h时比较差异均有统计意义(P<0.01);B~E组作用于AML细胞6h后均能诱导AML细胞凋亡,D组细胞凋亡率最高;2μmol/L雷公藤红素作用于AML细胞3,6,12,24h,其细胞凋亡率随时间延长而逐渐增高。结论雷公藤红素有明确抗AML细胞增殖的能力,其作用可能与诱导细胞凋亡有关。Objective To investigate the effects of tripterine on proliferation and apoptosis of acute myeloid leukemia primary cells in vitro.Methods The preliminary diagnosed acute myeloid leukemia cells were divided into five groups according to concentration of tripterine.Group A was blank group,and groups B to E were treated with 1,2,5 and 10 μmol/L tripterine.The effects of tripterine on proliferation and apoptosis were assayed with MTT and flow cytometry.Results Proliferation inhibiting ratio increased with the concentrations of tripterine after treating for different periods in groups B to E compared with group A(P0.01),there was a significant difference in groups C and D compared with group B(P0.01),and no significant difference compared with group E(P0.05).The optimal concentration of tripterine was 2 μmol/L.There were no significant differences in proliferation inhibiting ratio in 6-,12-and 24-hour treatment by tripterine(P0.05),which had significant differences with that of 3-hour treatment(P0.05).The acute myeloid leukemia cells apopsis was induced after treated for 6 hours in groups B to E,and apopsis rate was the highest in group D.After the treatment of 2 μmol/L tripterine for 3,6,12 and 24 hours,the apoptic rates increased with the time.Conclusion Tripterine possesses the potent of anti-acute myeloid leukemia primary cell proliferation,which may be associated with the apoptosis of acute myeloid leukemia cells.
