摘 要:目的了解口腔粘膜下纤维性变中胶原堆积的机制。方法对口腔粘膜角质形成细胞和成纤维细胞进行了共培养,并采用槟榔碱预处理角质形成细胞,用明胶酶谱方法检测其胶原酶的含量及活性。用碱水解法检测胶原含量。结果只有共同培养组才能检测到MMP-9含量;槟榔碱预处理共同培养组的活化型MMP-2水平明显升高(P<0.05)。单独培养组中无MMP-9表达。共同培养组中胶原水平明显低于单独培养组(P<0.05)。结论 OSF组织中胶原堆积可能与MMP-2,MMP-9的表达没有明显关系。[ Objective ] To explore the mechanism of collagen cumulating in oral submucous fibrosis. [Methods ] Oral keratinocytes were pretreated by arecoline and cocultured with fibroblasts. The level of collagen, the content and activity of matrix metalloproteinase were detected.[Results] MMP-9 was produced only by co-cultm'e group, The active form of MMP-2 in coculture groups were significantly higher than those of the non-cocuhure groups. The collagen concentration in cocuhure groups was significantly lower than that of non-coculture groups. [ Conclusion ] Collagen increasing in oral submucous fibrosis may not due to the expression of MMP.
