摘 要:目的研究在血小板源性生长因子作用下,大鼠血管平滑肌细胞表达膜型基质金属蛋白酶和基质金属蛋白酶组织抑制物的变化,同时探讨两者对基质金属蛋白酶-2活性的影响。方法体外培养的血管平滑肌细胞经血小板源性生长因子处理0 min、20 min和6 h后进行如下检测:利用明胶SDS-PAGE酶谱检测血管平滑肌细胞分泌基质金属蛋白酶-2活性;基因芯片和RT-PCR技术检测血管平滑肌细胞表达膜型基质金属蛋白酶和基质金属蛋白酶组织抑制物的变化。结果明胶酶谱测定表明:血小板源性生长因子显著提高血管平滑肌细胞分泌的基质金属蛋白酶-2的活性;基因芯片和RT-PCR结果显示:血小板源性生长因子对血管平滑肌细胞表达膜型基质金属蛋白酶有明显的诱导作用,作用20 min和6 h后的表达是对照组(0 min)的1.86倍和2.93倍(P<0.05);同样血小板源性生长因子显著增强基质金属蛋白酶组织抑制物的表达,作用20 min和6 h后的基因表达是对照组(0 min)的1.35倍和1.82倍(P<0.05)。结论血小板源性生长因子显著增强了血管平滑肌细胞对膜型基质金属蛋白酶和基质金属蛋白酶组织抑制物的表达,同时两者的协调作用使MMP-2的活性呈明显的增加趋势。[Objective] To investigate the expression of membrane type-1 matrix metalloproteinase (MT1-MMP) and tissue inhibitor of metalloproteinase-2 (TIMP-2) in vascular smooth muscle cells (VSMCs) treated with platelet- derived growth factor (PDGF). Furthermore to explore the synergetic effects of MT1-MMP and TIMP-2 on matrix metalloproteinasc-2 (MMP-2) activity. [ Methods ] After treatment with PDGF 0 rain, 20 rain and 6 hours, MMP-2 activity was assayed by gel zymography. Meanwhile the expression of MT1-MMP and TIMP-2 were screened by us- ing Chip and confirmed by RT-PCR. [Results] The gel zymography showed that the activity of MMP-2 was signifi- cantly improved by PDGF. The expression of MT1-MMP was increased 1.86-fold and 2.93-fold for 20 rain and 6 hrs treatment by PDGF compared with 0 min, respectively. And likewise the mRNA expressions of TIMP-2 were 1.35-fold and 1.82-fold higher in cultured with PDGF at 20 min and 6 hrs than 0rain, respectively. [ Conclusion ]PDGF significantly improved the expression of MT1-MMP and TIMP-2 in VSMCs and MMP-2 activity was enhanced by the coordination of MT1-MMP and TIMP-2.
