摘 要:目的探讨中心体γ-微管蛋白在人食管癌前病变及食管癌中的表达及其在食管癌发生发展过程中的作用。方法采用免疫组织化学法检测食管癌前病变及食管癌各40例中心体γ-微管蛋白表达水平,30例正常食管组织患者作为对照组,并用Western免疫印迹进行验证,所得数据进行统计学分析。结果不同样本组γ-微管蛋白表达水平不同(P=0.0001),由食管正常上皮组织到出现不典型增生至进展为食管癌发展变化,γ-微管蛋白表达量呈增高趋势,各组间差异均有统计学意义(P=0.001)。比较同例和同组γ-微管蛋白的表达程度差异无统计学意义(P〉0.05)。Western免疫印迹与免疫组化检测结果一致。结论中心体异常为食管癌细胞恶性表型的明显特征之一,并可能是食管癌形成过程中的早期事件。γ-微管蛋白表达情况可为食管癌发生机制的探讨、高危病例的筛选及早期诊断、早期治疗提供一种新的思路和途径。Objective To investigate the expression and the significance of γ- tubulin in premalignant lesion and esophageal squamous cell carcinoma tissues. Methods Forty specimens of premalignant lesion of esophageal epithelial tissues, 40 specimens of esophageal squamous cell carcinoma tissues, and 30 specimens of normal esophageal epithelial tissues were obtained during operation. Immunohistochemistry was used to analyze the protein expression of γ - tubulin. Western blotting was used to examine the mRNA expression of γ - tubulin. Results The protein and mRNA expression of γ- tubulin in esophageal squamous cell carcinoma tissues were higher than those in the premalignant lesion of esophageal epithelial tissues and normal esophageal epithelial tissues with significant differences between each other (P = 0. 001 ). There was no significant differences of the expression of γ - tubulin in same group (P 〉 0. 05). Conclusions squamous cell carcinoma. Aberration Centrosome protein is one of the distinct phenotypes of esophageal of centrosome proteins may be used to screen high risk cases and to investigate the development mechanism of the esophageal carcinoma and might become a new method to treat and prevent esophageal carcinoma.
