摘  要:目的:探讨喉鳞癌中缺氧诱导因子-1α(HIF-1α)的表达及其与肿瘤细胞凋亡和增殖的关系。方法:运用免疫组化二步法检测喉鳞癌组织中HIF-1α与增殖细胞核抗原的表达,同时用DNA末端标记法检测喉鳞癌原位细胞凋亡。结果:59例喉鳞癌组织标本中,HIF-1α阳性表达率为54.2%(32/59),显著高于喉部良性病变(P<0.05),并与肿瘤临床分期和淋巴结转移均显著相关(均P<0.05),与组织学分级无显著相关(P>0.05)。HIF-1α阳性表达组的增殖指数(55.50±16.37)显著高于阴性表达组(P<0.05),凋亡指数(3.01±1.19)低于阴性表达组,无显著性差异(P>0.05)。结论:HIF-1α在喉鳞癌的发生发展中起到一定的作用,并与肿瘤细胞的增殖有关,但与凋亡无关。Objective:To investigate the expression of hypoxia inducible factor-1α(HIF-1α)and its relationship to apoptosis and proliferation in laryngeal squamous cell carcinoma(LSCC).Methods:Immunohistochemical method was used to detect the expression of HIF-1α and proliferating cell nuclear antigen(PCNA).Tunel technique was used to detect in situ cell apoptosis in LSCC.Results:Fifty nine cases of LSCC samples,HIF-1α expression was 54.2%(32/59),significantly higher than benign laryngeal lesion.The expression of HIF-1α was related with clinical stages of cancer and lymph node metastasis(P0.05),but not related to histological grades(P0.05).PCNA-labeling index was higher in HIF-1α positive group(55.50±16.37)than that in negative group and the difference was statistically significant(P0.05).However,although it was lower of apptosis index in HIF-1α positive group(3.01±1.19)than that in negative group,the difference was found to have no statistical significance(P0.05).Conclusion:HIF-1α may play a role in the carcinogenesis of laryngeal carcinoma and is related with proliferation,but has no relationship with the apoptosis of tumor cells in LSCC.
