摘  要:目的探讨紫花牡荆素(Casticin,CAS)是否通过线粒体凋亡途径诱导人宫颈癌HeLa细胞凋亡。方法体外培养人宫颈癌HeLa细胞。ELISA法检测细胞组蛋白/DNA碎片;碘化丙啶(propidium iodide,PI)染色流式细胞术(flow cytometry,FCM)分析细胞凋亡率;比色法测定细胞半胱天冬酶caspase-9,-3活性;罗丹明123探针FCM检测细胞线粒体跨膜电位。结果 CAS以浓度依赖方式增高HeLa细胞组蛋白/DNA碎片水平和凋亡率(P<0.05)。CAS增强HeLa细胞caspase-9,-3活性(P<0.05)和诱导Hela细胞线粒体膜电位下降(P<0.05),呈浓度依赖性。结论 CAS诱导HeLa细胞凋亡涉及线粒体凋亡途径的激活。Objective To investigate whether casticin(CAS) induces apoptosis via activation of mitochondrial apoptotic pathway in human cervical cancer HeLa cell line. Methods Human cervical cancer HeLa cells were cultured in vitro. Histone/DNA fragment was examined using ELISA assay. Apoptotic rate was determined by flow cytometry(FCM) using propidium iodide staining. The activity of caspase-9,-3 was measured using colorimetry. The fluorescent probe rhoda- mine 123 (Rh 123) was used to analyze mitochondrial transmembrane potential and the cellular fluorescence intensity was measured by FACS. Results CAS significantly increased the level of Histone/DNA fragment and the percentage of apoptotic cells in HeLa cells (P 〈 0.05 ), in a concentration-dependent manner. CAS activated caspase-9 and -3 and reduced the mitochondrial transmembrane potential of HeLa cells (P 〈 0.05) ,in a concentration-dependent manner. Conclusion Induction of apoptosis of HeLa ceils by CAS treatment is involved in activation of mitochondrial apoptotic pathway.
