摘  要:为纯化树莓叶片黄酮成分,比较了7种树脂静态吸附和解吸过程,筛选出合适的大孔树脂初步纯化树莓叶片总黄酮,优化了吸附和解吸条件。结果表明:AB-8大孔树脂对树莓叶片总黄酮有较好的吸附和解吸性能,其最佳工艺为上样液pH=6.0、流速2.0 mL·min-1,洗脱剂乙醇最适宜浓度为70%,流速1.5 mL·min-1,经AB-8大孔树脂分离纯化后,总黄酮含量从3.46%增加到45.62%,提高了13.18倍。样品经硅胶和Sephadex LH-20凝胶色谱进一步分离纯化后得到2个黄酮苷元单体,通过HPLC、核磁共振分析,确定为槲皮素和山奈酚,纯度分别为98.6%,98.9%,在树莓叶片中含量分别为179.33μg·g-1、60.77μg·g-1(苷元含量)。In order to purify the flavonoids extracted from Raspberry leaves,the static adsorption and desorption behaviors of seven kinds of macroporous resins used for the separation of flavonoids were studied respectively.Among them,the most suitable kind of macroporous resin was screened for the preliminary purification of the flavonoids extracted from the Raspberry leaves,and its adsorption and desorption conditions were optimized too.The results show that the AB-8 macroporous resin offers the best adsorption and desorption behaviors for total flavonoids in the extractive of the Raspberry leaves.Using AB-6,the optimum purification conditions are as follows: sample pH is 6,sample flow rate is 2.0 mL·min-1,the elution used is alcohol with concentration 70% and the eluting rate is 1.5 mL·min-1.After the AB-8 resin treatment,the total flavonoids content in the product increases 13.18-fold from 3.46% to 45.62%;and then the silica gel separation and Sephadex LH-20 gel chromatography were used to further purify the flavonoids obtained.Two flavonoid aglycones monomers,quercetin and kaempferol,were obtained and the purities of them are 98.6% and 98.9%,respectively,with HPLC and NMR analyses,and their contents in the Raspberry leaves are 179.33 μg·g-1 and 60.77 μg·g-1(aglycone content),respectively.
