摘 要:目的探讨体外缺氧条件下姜黄素对人胰腺癌细胞PANC-1中缺氧诱导因子-1α(HIF-1α)的抑制作用和机制。方法缺氧条件下体外培养PANC-1,RT-PCR检测不同剂量的姜黄素作用24h后HIF-1αmRNA表达的变化;ELISA检测处理后PANC-1细胞中HIF-1α蛋白的表达水平。结果 PANC-1细胞在0.1、1、10、100μmol/L姜黄素处理后,HIF-1α蛋白水平与对照组比较明显降低(F=138.43,P<0.01),且有明显的剂量依赖性,随剂量的增大而降低,但各剂量组HIF-1α的mRNA水平与对照组比较无显著性差异。结论缺氧条件下,姜黄素抑制人胰腺癌PANC-1细胞HIF-1α的转录活性,是通过抑制HIF-1α蛋白的合成来调控的。Objective To investigate the inhibitory effect and mechanism of curcumin on induction of HIF-1α gene in human pancreatic carcinoma PANC-1 cells under in vitro hypoxia environment. Methods PANC-1 cultured in vitro was placed in an incubator with hypoxia for 24h. RT-PCR was used to examine the effect of different doses of curcumin on the expression of HIF-1α mRNA in PANC-I cells. Effect of curcumin on the expression of HIF-1α protein was studied by ELISA. Results PANC-I cells were disposed by curcumin with the dose of 0.1, 1, 10, 100μmol/L respectively, HIF-1α protein levels obviously decreased in comparison with control group and were dose- dependently suppressed with the increasing doses of curcumin. But no significant differences were found between the mRNA of HIF-1α, either in experimental group or control group. Conclusion Curcumin inhibits the expression of HIF-1α VEGF gene via down-regulation of HIF-1α protein.
