摘  要:目的探讨帕金森病(PD)大鼠行为学模型的实用性及黑质神经元的凋亡特征。方法注射6-羟基多巴胺(6-OHDA)制备大鼠模型,5周后用自制梯子和跑步机模型测试大鼠行为学指标。冰冻切片HE染色观察大鼠黑质神经元的形态特征,对比其凋亡情况。结果在梯子实验中,PD模型大鼠与正常对照组和假手术组大鼠相比,行走速度下降,行走时间延长(P<0.05),行走失误率显著增加(P<0.01);在跑步机实验中,PD模型大鼠较正常对照组持续爬行时间显著缩短,一定时间内的跌落次数增加(P<0.01)。组织学观察PD模型大鼠注射药物侧黑质神经元显著减少,凋亡细胞显著增多,胞质染色加深。结论本文造模大鼠在自制梯子和跑步机实验中的行为学改变与黑质神经元凋亡程度相一致,此实验方法可以初步用于PD大鼠模型的鉴定。Objective To assess the usefulness of Parkinson' s disease (PD) rats' behavior model, and discuss the relevance of PD rats' behavioral changes with substantia nigra neuronal apoptosis. Methods 6-OHDA was injected into the SNc and the VTA of the rats to produce PD models. After 4 weeks, the behavior tests including the ladder model and the treadmill model were measured, then HE staining was used to detect the rats' morphology of neurons in substantia nigra, in the same time, the apoptosis degree was compared. Results In the ladder experiment, compared with the sham group and the normal group, the walking speed of PD rats was compared decreased, the walking distance was prolonged, and the walking error rate was also increased (P 〈 0.05, 〈 0.01 ). In the treadmill experiment, the creeping time decreased, but the falling time was increased within a certain period (P 〈 0.01 ). Histologic observation, the substantia nigra neurons of the model rats' side which injected drugs was decreased, apoptotic cells was increased significantly and the staining of the cytoplasm was deepened. Conclusions The behavioral changes of model tats induced by 6-OHDA were associated with the substantia nigra' s neuronal apoptosis in both the treadmill test and the ladder test. This method can be used for preliminary identification of PD model.
