培养基不同血清比例对人胃癌SGC-7901细胞生长的影响

摘　　要：目的:探讨细胞培养基中不同血清比例对人胃癌细胞SGC-7901生长状态的影响,为减少培养该细胞时血清的用量。方法:用分别含有3%、6%和9%小牛血清的1640培养液分别培养SGC-7901细胞12h、24h和48h,以无血清培养基作为空白对照;倒置显微镜观察细胞的生长状况,通过基于酶标仪的MTT方法检测细胞在不同比例血清中的生长与增殖的差异。结果:SGC-7901细胞在含3%和6%小牛血清的细胞培养基中生长状态与常用的含9%小牛血清的培养基中的生长情况相比,未见显著性的差异(P>0.05);也不呈时间依赖性,但与空白血清对照组相比差异有统计学意义(P<0.05)。结论:在体外培养RPMI-1640细胞时,可将培养基中小牛血清的比例降至3%～6%,从而降低在培养该细胞过程中的的实验成本,减少不必要的血清浪费。Objective：Through the cultivation of influence that different percentages of serum infect on the human gastric cancer SGC-7901 cells,and difference which The human gastric cancer SGC-7901 cells growth on 3 kinds percentages of serum.Methods：The human gastric cancer SGC-7901 cells were treated with 3%,6%,9% of blood serum and group NH withou blood serum for 12 to 48 hours.Morphological changes of cells were observed with invert microscope and growth rates of SGC-7901 cells were measured with MTT method and Microplate.Results：Different percentage of blood serum and time-dependence have no influence on the cell growth.Conclusion：The human gastric cancer SGC-7901 cells can grow well in these 3 different percentage of blood serum,so we use 3% of the blood serum instead of the other two to reduce the cost of the experiments and serum.
