摘  要:目的研究脐血来源细胞因子诱导的杀伤(CIK)细胞表型变化与细胞毒活性相关性。方法用流式细胞术测定诱导前脐血单个核细胞及诱导后培养7、14和21 d CIK细胞的表型;用CCK-8测定培养0、7、14和21 d CIK细胞按20∶1、10∶1、5∶1与肿瘤细胞U251、SiHa和A549接种时对其的细胞毒活性。结果 CIK细胞对U251、SiHa和A549的杀瘤率,7、14和21 d依次降低(P<0.05);CIK细胞接种比例越高,杀瘤率越高(P<0.05);CIK细胞杀瘤率与CD3-CD19+、CD3+CD4+CD8-、CD3+CD45RA-CD45RO+、CD3+CD25-CD28+、CD3+CD25+CD28+呈正相关(P<0.05),与CD3+CD4-CD8-、CD3+CD16+CD56+呈负相关(P<0.05)。结论脐血CIK细胞对肿瘤细胞有很强的杀伤活性,且杀伤活性与CIK细胞各细胞表型及其与肿瘤细胞的比例有关。Objective To research the correlation between phenotypes of cytokine-inducted killer (CIK) cells from human umbilical blood and their eytotoxic activity. Methods The phenotypes of CIK cells cultured 0,7,14 and 21 d were detemfined by flow cytometry. And the cytotoxie activity of CIK cells to U251 ,SiHa and A549 with different ratio was detected with CCK-8 assay. Results The eytotoxic activity to U251, SiHa and A549 of CIK cells cultured 7 d was higher than that of 14 d,while that of 21 d was the lowest (P 〈 0.05 ) ;and the higher ra- 50 of CIK cells,the higher cytotoxic activity (P 〈 0.05). The cytotoxie activity of CIK cells was correlated positively with CD3- CD19 + , CD3 + CD4 + CD8 - , CD3 + CD45RA- CD45RO + , CD3 + CD25 - CD28 + , CD3 + CD25 + CD28 + ( P 〈 0.05 ), and negatively with CD3 + CD4 CD8- ,CD3 + CD16 + CD56 + (P 〈 0.05). Conclusion CIK cells from human umbilical blood have very intense cytotoxic activity to tumor cells, and the cytotoxic activity is correlated with the phenotypes of CIK cells and the ratio of these cells.
