摘 要:目的探讨纤溶酶对在体小鼠肺泡液体清除率(AFC)的作用。方法应用考马斯亮蓝法测定小牛血清白蛋白浓度的方法测定小鼠在体AFC。结果特异性上皮细胞钠通道阻断剂阿米洛利组能明显降低AFC,气管内注入60μg/mL纤溶酶后,AFC明显增加(38.2±2.1)%,与对照组相比P<0.05,n=6)。纤溶酶+阿米洛利组对AFC的影响与对照组相比差异无统计学意义(P>0.05,n=5),与单独使用纤溶酶比较差异有统计学意义(P<0.05)。结论纤溶酶能明显增加肺泡上皮液体的清除,可能通过增强阿米洛利敏感性钠通道活性而影响AFC。Objective Aim Compared with its wide usage in the treatment of pleural effusion preventing pleural ad- hesion and fibrosis, the mechanism of plasmin in the regulation of absorption of lung edema was unknown. We aim to ex plore the relationship between plasmin and alveolar fluid clearance (AFC) in mice in vim . Methods In vivo AFC was measured using bovine serum albumin protein assays by Coomassie brilliant blue method. Results The rate of in vivo AFC was significantly decreased in mice intratracheally administrated amiloride (a specific ENaC blocker), and 60μg/mL plas- min could increase AFC in mice significantly (38. 2+_2.1) %, P〈0.05 compared with control, n=6). In the presence of both plasmin and amiloride, fluid resolution showed no much difference with that of control group (P;〉0.05, ,l= 5), whereas much difference with that of plasmin group (P%I0. 05). Conclusion Plasmin could increase alveolar fluid resolu tion significantly, probably by enhancing epithelial sodium channel and affecting AFC.
