摘 要:目的分析骨肉瘤线粒体DNA(MtDNA)含量的变化,探讨线粒体DNA在骨肉瘤发生、发展中的作用。方法应用荧光定量PCR(quantitative real time PCR,QPCR)技术检测20例骨肉瘤组织以及5例正常骨组织MtDNA含量,并结合骨肉瘤主要临床病理参数进行分析。结果骨肉瘤组织中MtDNA含量较正常骨组织降低了71%,差异有统计学意义(P<0.05),且MtDNA含量与骨肉瘤的转移密切相关(P<0.05),而与年龄、性别、肿瘤部位以及肿瘤病理学类型均无相关性。结论 MtDNA含量降低与骨肉瘤的发生、发展有着密切关系,并有可能成为一种新的骨肉瘤转移性标记物。Purpose To explore the effect of mitochondrial DNA (MtDNA) in the initiation and progression of osteosarcoma by analyzing the alterations of MtDNA content in osteosarcoma tissue. Methods The MtDNA content in 20 osteosarcoma tissue and 5 normal bone controls was examined by quantitative real-time polymerase chain reaction analysis. Moreover, the possible correlations between MtDNA content and clinicopathological parameters in osteosarcoma were analyzed. Results MtDNA content was significantly decreased by 71% in cancerous specimens as compared to normal bone controls. There was statistical correlation between MtDNA content and tumor metastasis, while not with age, gender, tumor location, and pathological type. Conclusions The occurrence and development of osteosarcoma were associated with decreased MtDNA content, which could be a noval metastatic marker of osteosarcoma.
