针刺督脉对脑缺血再灌注大鼠神经细胞凋亡和Caspase-8蛋白表达的影响

摘　　要：目的:研究针刺督脉经穴对局灶性脑缺血大鼠脑细胞凋亡和Caspase-8表达的调节作用,探讨针刺督脉经穴对脑缺血的脑保护机制。方法:线栓法制作雄性SD大鼠大脑中动脉栓塞(MCAO)模型。通过TUNEL、免疫组化法观察各组针刺后细胞细胞凋亡和Caspase-8表达的变化。结果:大鼠MCAO后大量细胞凋亡,Caspase-8表达增加,针刺督脉组MCAO 24 h后凋亡细胞数减少,24 h和72 h Caspase-8阳性细胞数降低。结论:针刺督脉经穴可减少细胞凋亡,抑制Caspase-8的过度表达,这可能是针刺督脉经穴治疗缺血性脑损伤的作用机制之一。Objective：To investigate the effect of acupuncturing Governor Vessel on neuron apoptosis and Caspase-8 expression for ischemic cerebrovascular disease in rats.Methods：The MCAO rats were prepared by suture method.The neuron apoptosis and Caspase-8 expression were determined by TUNEL and immunohistochemical method.Results：The neuron apoptosis and Caspase-8 expression were significantly higher after MCAO.The positive cells of neuron apoptosis at 24 h after MCAO in therapy group were significantly decreased.The levels of Caspase-8 expression at 24 h and 72 h after MCAO in therapy group were decreased.Conclusion：Acupuncturing Governor Vessel may reduce neuron apoptosis and caspase-8 expression in MCAO rats.It may be the mechanism of acupuncturing on Governor Vessel therapy to ischemic cerebrovascular disease.
