摘  要:目的通过RNA干扰技术阻断卵巢癌SKOV3细胞中AKT基因的表达,研究其对卵巢癌细胞增殖与凋亡的影响。方法人卵巢癌SKOV3细胞体外培养,分为对照组、空白载体组与RNA干扰组三组,采用MTT法与流式细胞仪AnnexinV/PI法检测并评价AKT基因干扰后对卵巢癌细胞增殖、凋亡的影响。结果 MTT结果显示RNA干扰组细胞增殖能力较对照组与空白载体组明显降低,差异具有统计学意义(P<0.05),空白载体组与对照组比较差异无统计学意义(P>0.05);流式细胞仪AnnexinV/PI法检测结果显示,RNA干扰组细胞凋亡率高达到(79.52±2.31)%,较对照组与空白载体组明显升高,约是空白载体组与对照组的3倍,差别具有统计学意义(P<0.05)。结论靶向AKT基因的小干扰RNA(siRNA)可有效地抑制卵巢癌细胞的增殖,促进细胞凋亡,是卵巢癌的治疗新方法。Objective To inhibit the key gene AKT of SKOV3 ceils by RNA interference and study the effect of RNA interference on ovarian cancer cell proliferation and apoptosis. Methods Human ovarian carcinoma SKOV3 cells in vitro were divided into the control group, the empty vector group and the RNA interference group. The ovarian cancer cell proliferation, apoptosis was evaluated by MTT method and flow cytometry AnnexinV/PI assay in each group. Results MTT results showed that the cell proliferation in the RNA interference group was significantly lower as compared with the control group and the empty vector group ( P 〈 0. 05 ). But there was no significant difference in the cell proliferation between the empty vector group and the control group ( P 〉 0.05 ). The flow eytometry AnnexinV/PI assay showed that the apoptosis rate of RNA inter- ference group ( (79.52 ± 2. 31 ) % ) was significantly higher than that of the empty vector ( P 〈 0. 05 ) and was about 3 times of the empty vector group and the control group. Conclusion AKT - gene - targeted siRNA could inhibite cell proliferation and promoted the apoptosis of ovarian cancer cells, which was a new effective treatment method for ovarian cancer.
