摘  要:目的研究甲基化抑制剂5-氮杂-2'-脱氧胞苷(5-Aza-CdR)对人肾痛A498细胞增殖及γ-连环蛋白(γ-catenin)表达的影响。方法使用不同浓度(10^(-7)、10^(-6)、10^(-5)mol/L)的特异性DNA甲基转移酶抑制剂5-Aza-CdR处理A498肾癌细胞株。MTT检测5-Aza-CdR对肾癌细胞株A498抑制作用。细胞免疫化学检测5-Aza-CdR处理A498肾癌细胞株后γ-catenin表达的变化。结果 5-Aza-CdR能明显抑制肾癌细胞的增殖,且与药物浓度及作用时间有关。药物处理后y-连环蛋白表达增加。结论 y-catenin蛋白表达受甲基化的抑制,提示5-Aza-CdR可使γ-catenm基因去甲基化而抑制A498肾癌细胞株增殖。Objective To investigate the effect of methylation inhibitor 5 - Aza - 2' - deoxycytidine on the growth of human renal cancer cell line A498 and γ- catenin expression. Methods Human renal carcinoma A498 ceUs were treated with different concentrations of 5 - Aza - CdR( 10 -7,10 -6,10 -5 mol/L) respecttively. Then the growth rate of the ceils was detected by MTT assay. The immuneocytochemistry was used to detect γ - catenin levels in the cell line before and after treatment with 5 - Aza - CdR. Results 5 - Aza - CdR can inhibit the growth of A498 cells, 7 - catenin expression levels in A498 ceils were increased after 5 - Aza - CdR treatment. Conclusion γ - catenin expression levels were inhibited by methylation, 5 - Aza - CdR can inhibit the growth of A498 cells by inducing demethylation in the promoter region of γ - catenin gene.
