摘 要:目的观察中介素1-53(IMD1-53)对脂多糖(LPS)致急性肺损伤(ALI)大鼠体内白介素(8IL-8)和白介素2(3IL-23)表达的影响。方法将健康雄性Wistar大鼠48只随机分为正常对照组(C组),LPS致ALI组(ALI组),IMD1-53干预组(D组)。尾静脉注射脂多糖5mg/kg制作大鼠急性肺损伤模型。各组于2h、4h两点各组处死8只大鼠取血浆和右肺,ELISA法测血清IL-23水平;免疫组化法测组织中IL-8的水平,观察各组肺脏湿/干质量比值(W/D),观察各组肺组织病理形态学变化。结果与ALI组相比,中介素1-53干预组能显著降低IL-8和IL-23的表达,病理变化及肺脏湿/干质量比值显示中介素1-53对ALI有保护作用。结论 IMD1-53对大鼠急性肺损伤有保护作用,该保护作用可能是通过降低IL-8、IL-23等炎性因子的表达实现的。Objective To investigate the effect of intremedin 1-53 on IL-8 and IL-23 expression in rats with acute lung injury induced by lipopolysaccharide. Methods Forty -eight healthy Wistar rats were randomly divided into three groups:normal controls group(C group), acute lung injury induced by lipopolysaccharide group(ALI group)and intermedinl-53-pretreated group(D group). 5mg/kg intravenous injection of lipopolysaccharide rat model of acute lung injury.At 2h, 4h two time points ,8 rats from each group were killed and lung plasma obtained, Measured by ELISA serum levels of IL-23; immtmohistochemical method to measure tissue levels of IL-8, observed in each group lung wet / dry weight ratio (W / D), To observe the morphological changes in lung tissue. Results Compared with the ALl group, intermedin 1-53 in the intervention group significantly reduced IL-8 and IL-23 expression, pathological changes and lung wet / dry weight ratio of the intermedinl-53 shows a protective effect on ALI. Conclusion IMD1-53 has a protective role in acute lung injury in rats, the protective effect may be through reduced IL-8, IL-23 expression of inflammatory factors such as implementation.
