酪酸梭菌联合美沙拉嗪对溃疡性结肠炎小鼠的治疗作用及其机制

摘　　要：目的观察酪酸梭菌联合美沙拉嗪对小鼠溃疡性结肠炎(UC)的治疗作用,并探讨其可能的作用机制。方法用3%的葡聚糖硫酸钠(DSS)建立小鼠溃疡性结肠炎模型。50只BALB/c小鼠随机分为正常组、模型组、酪酸梭菌组、美沙拉嗪组、酪酸梭菌联合美沙拉嗪组。观察指标包括疾病活动指数(DAI)评分,结肠粘膜肉眼改变及病理组织学积分,采用免疫组化法检测结肠粘膜组织中HSP27的表达量,ELASA法检测血清中TNF-α的表达量。结果酪酸梭菌和美沙拉嗪都可以降低实验小鼠DAI积分,减轻肠道的组织学损伤程度;与模型组相比,酪酸梭菌组、美沙拉嗪组、酪酸梭菌联合美沙拉嗪组,结肠中HSP27的表达都上调(P<0.05),血清中TNF-α的表达都下降(P<0.05),其中美沙拉嗪组与酪酸梭菌组治疗效果相当(P>0.05),酪酸梭菌联合美沙拉嗪组治疗效果最佳(P<0.05)。结论酪酸梭菌和美沙拉嗪对急性期UC都有治疗作用,二者联合用药效果更佳,其部分机制可能与上调HSP27,抑制TNF-α的表达而减轻肠道的炎性反应有关。Objective To investigate the effects of clostridium butyricum combined with mesalazine on ulcerative colitis in mice to find its possible mechanism of treating UC. Methods 3% dextran sulphate sodium（DSS） was used to establish experimental UC model in BALB/c mice. Fifty mice were randomly divided into five groups as follows：normal control group,model control group, mesalazine group ,clostridium butyricum group, clostridium butyricum combined with mesalazine group. Disease activity index（DAD and histological injury scores were used to assess the efficacy of intervention groups. The expression of HSP27 in colonic mucosa were measured by immunohistochemistry（IHC） and TNF-α in serum by ELASA method. Results Either clostridium butyricum or mesalazine could reduse the DAI and histological injury scores. Compared with model control group,in three treatment group , the expression of HSP27 in the colonic mucosa was increased（P〈0.05） and the expression of TNF-α in the serum wase decreased（P〈0.05）. Conclution Both clostridium butyricum and mesalazine had therapeutic effects on the acute U C mice. Clostridium butyrieum combined with mesalazine was more effective. Its possible mechanism may be related to up-regulating of HSP27 expression and inhibition of TNF-α.
