摘 要:为优化超声波法和酶解法提取菊花总黄酮的工艺,并比较其工艺条件,选择因素水平,采用L9(34)正交设计优化提取工艺,以芦丁为对照品,用紫外分光光度计分析其提取率.结果表明,酶解法最佳工艺条件为:25倍70%的乙醇溶液,酶添加量0.5%,于45℃水浴下酶解150min,得率为5.60%;超声波法最佳工艺条件为:10倍70%的乙醇溶液超声40min,提取3次,得率为4.61%.酶解法的得率比超声波法的高21.5%.Objective To optimize the Ultrasonic Extraction and Enzymatic Hydrolysis Extraction, and to compare these two methods. Methods With the Rutin as the reference substance and ultraviolet spectro- photometer for analyzing the extraction rate, the extraction process is optimized after the level of factor is chosen and an orthogonal designing method L9 (3^4) is adopted. Results The superior process conditions for the Enzymatic Hydrolysis Extraction are. 25 Ethanol solution (70%), with cellulose (0.5%), constant temperature (45 ℃), water bath for 150 minutes. Extraction rate is 5.60%. The superior process conditions for the Ultrasonic Extraction are: 10 Ethanol solutions (70%), ultrasound for 40 minutes, extract for 3 times. Extraction rate is 4.61%. Conclusion The extraction rate by the Enzymatic Hydrolysis Extraction is 21.5% higher than the rate by the Ultrasonid Extraction.
