不同葡萄品种CBF_1基因的克隆及序列分析

摘　　要：根据欧洲葡萄CBF1基因序列设计1对特异引物,采用PCR方法从山葡萄、贝达、SO4、Ln33和黑比诺的基因组中各扩增出1个DNA片段并克隆到pMD18-T载体中。经序列测定和分析,各片段均长756 bp,与欧洲葡萄CBF1基因的核苷酸序列及其推导的氨基酸序列分别具有98%和99%的同源性,而且推导的氨基酸序列中包含有同源性更高的AP2/EREBP DNA结合域,以及CBFs蛋白的两段特征序列(PKK/RPAGRxKFxETRHP和DSAWR)。结果表明从5种葡萄中均可以克隆出CBF1基因。A DNA fragment was cloned from the genome of 5 varieties of grape,Vitis amurensis,Beta,Ln33,SO4 and Pinot noir by polymerase chain reaction（PCR）using the primers designed following Vitis vinifera CBF1 genes.Sequence analysis showed that the 5 cloned DNA fragments all were 756 bp and 98 % and 99% identical to the Vitis vinifera CBF1 genes in nucleotide and deduced amino acid sequences respectively.Moreover,they showed one AP2/EREBP DNA binding domain and two signature sequences of the CBFs family proteins,PKK/RPAGRxKFxETRHP and DSAWR in the deduced amino acid sequence.
