MMP-9、Runx3蛋白在十二指肠腺癌中的表达及其临床意义

摘　　要：目的检测十二指肠腺癌中基质金属蛋白酶-9(MMP-9)及runt相关转录因子3(Runx3)蛋白的表达情况,分析其在十二指肠腺癌组织中表达与临床病理参数之间的关系。方法采用免疫组化法检测MMP-9及Runx3蛋白在十二指肠腺癌及癌旁正常组织的表达情况。结果十二指肠腺癌组织中MMP-9阳性率(70.0%),明显高于癌旁正常组织(27.5%),差异有统计学意义(P<0.01);Runx3蛋白在癌组织中阳性率(23.3%)明显低于癌旁正常组织(77.5%),差异有统计学意义(P<0.01);十二指肠腺癌组织中表达MMP-9与浸润深度、分化程度及淋巴结转移有关(P<0.05);Runx3蛋白在癌组织中表达与分化程度及淋巴结转移有关(P<0.05);二者在十二指肠腺癌组织中表达存在明显的相关性(r=0.311,P=0.011)。结论 MMP-9及Runx3蛋白表达异常与十二指肠腺癌的发生和发展密切相关,检测十二指肠腺癌中两者的表达可作为临床预测患者的转移及预后情况的依据。Objective To investigate the expression of MMP-9 and runt related transcription factor 3（Runx3） protein and its possible relationship with the clinicopathological features of duodenal adenocarcinoma.Methods The expression of MMP-9 and Runx3 protein were examined in duodenal adenocarcinoma and normal tissues by immunohistochemical SP technique.Results The positive expression rate of MMP-9 protein in duodenal adenocarcinoma was 70.0%,which was significantly higher than that in normal tissues 27.5%（P〈0.01）.The positive expression rate of Runx3 protein in duodenal adenocarcinoma was 23.3%,which was significantly lower than that in normal tissues 77.5%（P〈0.01）.The expression of MMP-9 protein was correlated with infiltration,differentiation and lymph node metastasis（P〈0.05）.The expression of Runx3 protein was correlated with differentiation and lymph node metastasis（P〈0.5）,but negative correlation between the expression of MMP-9 and Runx3 proteins in duodenal adenocarcinoma（r=0.311,P=0.011）.Conclusion The expression of MMP-9,Runx3 protein are correlated with the development of duodenal adenocarcinoma.Detection of the expression of MMP-9 and Runx3 protein in duodenal adenocarcinoma can predict the metastasis and prognosis,and provide the basis for clinical targeted therapy.
