天花粉蛋白对IL-1β介导下的人牙周膜细胞MMP-1表达的影响

摘　　要：目的观察天花粉蛋白(TCS)对白细胞介素-1β(IL-1β)介导下人牙周膜细胞基质金属蛋白酶-1(MMP-1)表达的影响,为进一步研究对抗牙根吸收的可能途径提供实验依据。方法用IL-1β10 ng/ml作用于体外培养的人牙周膜细胞(hPDLCs),通过噻唑蓝比色测定法(MTT)及逆转录聚合酶链反应(RT-PCR),观察IL-1β对hPDLCs生长状态及MMP-1表达影响;将TCS 0.05 ng/ml作用于IL-1β介导状态下的hPDLCs,观察hPDLCs生长状态及MMP-1表达的变化。结果在IL-1β及IL-1β复合TCS作用下,hPDLCs的增殖状况无明显改变(P>0.05);IL-1β介导下,hPDLCs中MMP-1的mRNA表达呈上升趋势。相对于IL-1β单独作用,IL-1β复合TCS作用下,hPDLCs中MMP-1的mRNA表达下调。结论IL-1β介导下的hPDLCs的MMP-1表达增高;TCS有抑制IL-1β介导下的hPDLCs MMP-1表达增高作用。Objective To observe the effects of trichosanthin （TCS） on the expression of matrix metalloproteinase - 1 （ MMP - 1 ） in human periodontal ligament cells （hPDLCs） mediated by interleukin-1β （ IL -1β ）, and to provide experimental evidences for further research on possible methods of agonistic root resorption. Methods HPDLCs cultured in vitro were incubated within IL - 1β （ 10 ng/ml）. The effects of IL -1β on the growth condition and MMP - 1 expression of hPDLCs were observed by the methods of MMT colorimetry and RT - polymerase chain reaction. The hPDLCs mediated by IL -1β were treated with TCS（0. 05 ng/ml）, and the growth condition and MMP - 1 expression changes of hPDLCs were observed. Results Treated with IL - 1β or IL - 1β combined with TCS, there was no obvious change in the proliferation condition of hPDLCs （ P 〉 0.05 ）. The mRNA expression of MMP - 1 was upgraded significantly in hPDLCs mediated by IL - 1β. The mRNA expression of MMP - 1 was down - regulated in hPDLCs mediated by IL - 1β combined with TCS, compared with that of the hPDLCs mediated by IL - 1β alone. Conclusion The MMP - 1 expression increases in hPDLCs mediated by IL - 1β. TCS can inhibit the increase of MMP - 1 expression in hPDLCs mediated by IL - 1β.
