摘  要:目的探讨HSP70-HBsAg嵌合基因修饰DC瘤苗对肝癌动物模型的免疫治疗效果。方法用重组腺病毒介导HSP70-HBsAg基因修饰树突状细胞构建HSP70-HBsAg-DC瘤苗后,采用皮下注射、静脉注射和瘤体注射三种途径向荷瘤小鼠回输HSP70-HBsAg-DC瘤苗,比较观察HSP70-HBsAg-DC瘤苗对实验性肝癌的治疗效果和免疫保护作用。结果皮下注射HSP70-HBsAg-DC瘤苗治疗效果明显优于瘤体内注射、尾静脉注射(P<0.05),HSP70-HBsAg-DC瘤苗组肿瘤的生长速度明显慢于对照组(P<0.05)。结论皮下注射HSP70-HBsAg-DC瘤苗可能是实验性肝癌最佳的免疫途径。HSP70-HBsAg-DC瘤苗对肝癌荷瘤小鼠产生有明显的免疫治疗作用,能够诱导机体产生较强的抗肝癌免疫保护作用。Objective To observe the immunotherapy function for liver cancer animal model by HSP70-HBsAg gene-modified DC tumorvaccines, Methods DC tumor vaccines were prepared with recombinant adenoviruses carrying HSPT0-HBsAg chimeric gene. The HSP70-HBsAgDC tumor vaccines were injected by three ways: hypo-dermal, intra-vein and intra-tumors. The immunotherapy functions of DC tumor vaccines were observed and compared among three different injection ways. Results Subcutaneous injection HSP70-HBsAg-DC tumor vaccines were obviously better than the intra-vein and intra-tumors injection (P〈0.05). The growth velocity of tumor in HSP70-HBsAg-DC tumor vaccines group was significantly slower than in control group (P〈0.05). Conclusion Subcutaneous injection HSP70.HBsAg-DC tumor vaccines may be the best way of immunity. HSP70-HBsAg-DC tumor vaccines have an obvious immune therapy effect for liver cancer mice model and can induce strong immune protection to liver cancer
