摘 要:应用rDNA-ITS2序列,采用邻接法(NJ)、最大简约法(MP)和最大似然法(ML),以按蚊亚属Anopheles的中华按蚊An.(An.)sinensis和赫坎按蚊An.(An.)hyrcanus为外群,对采自中国的按蚊属Anopheles塞蚊亚属Cellia21种蚊进行了系统发育分析。结果表明:rDNA-ITS2序列的长度范围为435~838bp,塞蚊亚属蚊种间遗传距离为0.027~0.496,平均值为0.336。各种方法重建的系统发育树均显示新迈蚊系Neomyzomyia为单系,新塞蚊系Neocellia和迈蚊系Myzomyia为并系。21种蚊在系统发育树中均分为4支,伪威氏按蚊An.pseudowillmori和塞沃按蚊An.sawadwongporni未归入多斑按蚊种团(An.maculatus group),其它各种团和复合体成员种的聚类关系与形态分类结果一致。本研究重建的分子系统发育树阐明了中国按蚊属塞蚊亚属各系及蚊种之间的亲缘关系,ITS2序列具有足够的信息贡献量。The second internal transcribed space of ribosomal DNA (rDNA-ITS2) for 21 species in subgenus Cellia and 2 species in the subgenus Anopheles of the genus Anopheles were sequenced and analyzed. The trees of neighbor joining, maximum parsimony and maximum likelihood for these species were constructed using An. (An.) sinensis and An. (An.) hyrcanus as the outgroups. The range of ITS2 length was from 435 bp to 838 bp, and the level of pairwise genetic distances (p distance) among Cellia mosquitoes was from 0.027 to 0.496 (mean as 0.336). All the phylogeny trees showed that Neomyzomyia series was monophyletic; the Myzomyia series and Neocellia series were paraphyletic. Twenty-one species in the subgenus Cellia in China were clustered into 4 major clades, and the members of each series and group were respectively clustered together and supported by the morphology, except for An. (Cellia) pseudowillmori and An. (Cellia) sawadwongporni which did not fall into the maculatus group. This molecular phylogeny objectively shows the interspecific genetic relationships among the Chinese subgenus Cellia mosquitoes. The rDNA-ITS2 sequences are sufficient contributions to reconstruct the phylogeny of species in the subgenus Cellia.
