青蒿琥酯诱导垂体泌乳素瘤细胞凋亡并下调泌乳素蛋白表达

摘　　要：目的初步研究青蒿琥酯(artesunate,ART)对泌乳素瘤细胞及其泌乳素(prolactin,PRL)蛋白表达的作用效果。方法MTT观察青蒿琥酯作用细胞后,细胞增殖情况,并计算药物的半数抑制浓度(IC50),Hoechst33342染色观察细胞凋亡小体,TUNEL计算细胞凋亡率,Western blot和RT-PCR观察不同浓度药物作用细胞后,激素蛋白和其mR-NA的表达量。结果青蒿琥酯能有效抑制泌乳素瘤细胞的增殖,且这种抑制作用具有浓度依赖性,Hoechst33342染色发现,药物组对比对照组,出现大量凋亡小体,初步判断该药可诱导泌乳素细胞凋亡,Western blot和RT-PCR显示药物作用后,泌乳素蛋白及其mRNA表达下调。结论青蒿琥酯能有效的抑制垂体腺瘤细胞增殖,诱导细胞凋亡,下调激素分泌。Objective To observe the effect of artesunate（ART） on prolactinoma cells.Methods The rat pituitary adenoma cell line（MMQ） was chosed as in vitro cell model.MTT method was used to investigate the growth of MMQ cell after treated by ART.Hoechst33342 and TUNEL assaies used to observe apoptotic body and apoptotic rate.The levels of PRL and PRLmRNA after ART treatment were investigated by Western blot and RT-PCR.Results ART inhibited the growth of MMQ cell.Contrast with the control group,Hoechst 33342 assay showed that there were many apoptotic bodies.ART reduced the expressions of PRL and PRLmRNA,which were observed by Western blot and RT-PCR.Conclusion ART can induce apoptosis of MMQ and reduce the secretion of PRL.
