摘 要:目的观察全反式维甲酸(ATRA)对甲状腺鳞癌细胞株SW579细胞CDK4、CyclinD1、Rb表达的影响,以探讨其抑癌机制。方法分别以终浓度为10-7、10-6、5×10-6、10-5mol/L的ATRA作用SW579细胞株,对照组加入5μl无水乙醇,各组细胞均在培养24 h后加药。继续培养48 h,RT-PCR、Western blot检测SW579细胞CDK4、CyclinD1、Rb mRNA及其蛋白的表达。结果 ATRA作用后,RT-PCR检测结果表明,经不同浓度ATRA处理的细胞CDK4、Rb mRNA表达水平没有明显变化;CyclinD1的表达明显下调;Western blot检测结果表明经不同浓度ATRA处理的细胞CDK4蛋白表达水平没有明显变化,pRb蛋白的磷酸化水平明显下降,CyclinD1蛋白表达水平明显下降。结论 ATRA在一定浓度范围内可能通过下调甲状腺鳞癌细胞株SW579细胞CyclinD1的表达及Rb蛋白的磷酸化水平抑制细胞增殖。Objective To observe the effects of all-trans retinoic acid(ATRA) on the expression of CDK4,CyclinD1 and Rb gene of thyroid squamous cell carcinoma cell line(SW579) so as to explore its mechanism.Methods The cells were randomly divided into control group and ATRA groups treated with different dose of ATRA(final concentration of 10-7,10-6,5×10-6,10-5 mol/L).After 24 hours,5 μl absolute alcohol and the different concentrations of ATRA were added.After 48 hours,CDK4,CyclinD1 and Rb mRNA expression levels were determined by RT-PCR.CDK4,CyclinD1 and pRb protein expression levels were determined by Western blot.Results After cells were treated with ATRA,the mRNA expression of CDK4 and pRb were not increased;the mRNA expression of CyclinD1 was descreased;distinct changes in the expression of CDK4 protein was not observed;the expression of CyclinD1 protein was descreased;the level of pRb protein phosphorylation was descreased.Conclusions ATRA can inhibit the proliferation of thyroid squmaous cell carcinoma cell line in a dose-dependent manner.The mechanism may be related with decreasing of CyclinD1,pRb mRNA and protein expressions.
