HER2、PTEN在胃癌组织中的表达及意义

摘　　要：目的探讨人类表皮生长因子受体2(HER2)和第10号染色体同源缺失性磷酸酶-张力蛋白基因(PTEN)在胃癌组织中的表达及其与临床病理特征的关系。方法收集有效胃癌组织蜡块407例,在胃癌组织芯片上,应用免疫组化EnVision法检测HER2和PTEN蛋白表达水平,其结果与30例正常胃黏膜组织比较。分析HER2和PTEN在胃癌组织的表达与患者临床病理特征的关系。结果 HER2在胃癌组织中的阳性表达率为9.1%;PTEN在胃癌中的阳性表达率为20.1%,显著低于正常胃黏膜组织的100%(P<0.01)。HER2的表达与肿瘤分化程度、组织学Lauren′s分型相关(P<0.05);PTEN的表达与组织分化程度、组织学分型、淋巴结转移相关(P<0.05);HER2与PTEN的表达呈负相关(r=-0.344,P<0.01)。结论 PTEN蛋白的表达与胃癌的发生、发展密切相关;同时检测胃癌组织中的HER2和PTEN蛋白的表达对判断肿瘤的恶性程度和预后可能有一定意义。Objective To evaluate the expression and significance of human epidermal growth factor receptor 2（HER2） and phosphatase and tensin homolog deleted on chromosome ten（PTEN） in human gastric carcinoma tissues.Methods The expression levels of HER2 and PTEN protein in 407 human gastric carcinoma tissues were measured by EnVision immunohistochemistry method in tissues-microarry.The results were compared to those in 30 samples of normal gastric mucosa tissues.The relationships between their expressions and clinicopathological features were analyzed.ResultsPositive expression rate of HER2 was 9.1%.Positive expression rate of PTEN was 20.1%,which was significantly lower than 100% in normal gastric mucosa（P0.01）.The expression of HER2 was significantly correlated with differentiation and Lauren′s classification（P0.05）,which of PTEN was significantly correlated with differentiation,Lauren′s classification and lymphnode metastasis of gastric carcinoma（P0.05）.The expression of HER2 was negatively correlated with that of PTEN（r=-0.344,P0.01）.Conclusion The expression of PTEN is closely related with occurrence and development of gastric carcinoma.Combined detection of HER2 and PTEN proteins may benefit to evaluate the malignancy and prognosis of gastric cancer.
