摘 要:乳腺癌是妇女常见的恶性肿瘤之一,内分泌治疗是乳腺癌内科治疗的主要手段之一,但仅33%~50%的乳腺癌患者内分泌治疗有效,造成治疗失败的原因可能与癌组织局部高水平的雌激素有关,其产生的重要原因是肿瘤组织自身合成的雌激素,在乳腺癌发生和发展中作用可能比外周血中的雌激素更为重要。在乳腺癌中主要有两个途径影响雌激素的局部合成,即类固醇硫酸酯酶(steroid sulfatase,STS)和芳香化酶(aromatase,AROM)途径。类固醇硫酸酯酶和雌激素硫酸转移酶(estrogen sulfotransferase,EST)是硫酸酯酶途径雌激素合成与代谢的两个重要酶,芳香化酶是芳香化酶途径的重要酶。本文对STS、EST、AROM在乳腺癌内分泌治疗中的研究做一综述。Breast cancer is a malignant tumor, which can be commonly found in Iemales. Hormonal treatment has become one of the major therapies for breast cancer, however only 33% to 50% of breast cancer cases can effectively respond to hormonal treatment. The failure in treatment could be related to the high concentration of estrogens in the tumor. Local biosynthesis of estrogens is believed to play a very important role in the high concentration of estrogen in breast carcinoma and may be more important than circulating estrogens to the occurrence and development of breast cancer. Local biosynthesis of estrogens is mainly influenced by two pathways in breast carcinoma, namely, sulfatase and aromatase pathways. Steroid Sulfatase ( STS ) and Estrogen Sulfotransferase ( EST ) are two significant enzymes that are associated with the synthesis and metabolism of estrogen in the sulfatase pathway, whereas Aromatase ( AROM ) is important in aromatase pathway. The current paper reviews the study of hormonal treatment for breast carcinoma.
