HSG、MMP-2和MMP-9在乳腺癌组织中的表达及意义

摘　　要：[目的]探讨新的增殖抑制基因(hyperplasic suppressor gene,HSG)和基质金属蛋白酶-2、9(matrixmet-alloproteinase-2,-9MMP-2,9)在乳腺癌组织中的表达及意义。[方法]应用免疫组化方法检测唐山市工人医院52例乳腺癌标本和18例乳腺纤维腺瘤标本中HSG、MMP-2和MMP-9表达。[结果]52例乳腺癌组织中HSG、MMP-2和MMP-9的表达分别为46.2%(24/52)、73.1%(38/52)和63.5%(33/52),在乳腺纤维腺瘤中,MMP-2和MMP-9着色很少,HSG的着色很深,表明三者与肿瘤的发生有关;MMP-2的表达与肿瘤的TNM分期、淋巴结转移和预后复发有关(P﹤0.05);乳腺癌组HSG低表达和MMP-9高表达,与淋巴结转移有关(P﹤0.05),且HSG和MMP-2、MMP-9呈负相关(r=-0.27,P﹤0.05)。[结论]乳腺癌组织中MMP-2高表达预示着病人预后更差,比MMP-9对乳腺癌与后的判断更有价值。综合检测HSG、MMP-2和MMP-9的表达更有利于了解乳腺癌转移的作用机制,更好地判断乳腺癌的进展及预后。[Objective]To determine the expression and significance of the novel hyperplasic suppress gene（HSG）and matrix metalloproteinase-2,-9（MMP-2,-9）in the process of breast cancer.[Methods]Immunohistochemistry method was used to measure the expression of HSG and metalloproteinase-2,-9,in which 52 cases were breast cancer samples,18 cases were breast fibroadenoma samples.All these cases were collected by the Department of pathology of the Tangshan Worker/s Hospital.[Results]The positive rate of HSG,MMP-2,-9 was 46.2%（24/52）,73.1%（38/52）and 65.3%（33/52）in breast cancer tissues,respectively.A very weak staining of MMP-2,-9 was observed in only a few epithelia in surrounding background breast tissue and fibrooadenoma tissue,but observed a very strong staining of HSG.The presence of HSG,MMP-2,-9 was significantly greater in breast cancer tissues compared with all other breast tissues;The high leve of MMP-2 was associated with the tumor grade,lymph node metastasis and increased recurrence of breast cancer（P﹤0.05）.The high level MMP-9 and the low level of HSG correlate with lymph node metastasis（P﹤0.05）,and HSG was negatively link to MMP-2 and MMP-9（r =-0.27,P﹤0.05）.[Conclusion]The study suggests that high expression of MMP-2 in breast cancer tissue may be associated with an unfavorable prognosis.
