摘 要:目的:研究溶血凝脂酸受体(LPAR)的表达与卵巢癌转移的关系。方法:小室细胞侵袭试验检测13种卵巢癌细胞的转移能力;实时定量RT-PCR检测各细胞株、10例正常卵巢组织、32例上皮性卵巢癌中LPAR1、LPAR2及LPAR3mRNA的表达。结果:有7种卵巢细胞可被LPA诱导迁移,为侵袭性细胞;另6种对LPA反应差,为非侵袭性细胞。侵袭性细胞中均有LPAR1的表达,显著高于非侵袭性细胞(P=0.003),LPAR2及LPAR3表达与癌细胞的侵袭能力无相关性。3种受体中,仅LPAR2在卵巢癌中的表达显著高于正常卵巢组织(P=0.030)。卵巢癌中,LPAR1的表达与临床分期有关,期别越高表达越高(P=0.032);LPAR2的表达与病理分级有关,分化越差表达越高(P=0.020)。存在广泛腹腔转移的病例均有LPAR1的表达。结论:LPAR1是介导LPA诱导的卵巢癌转移的主要受体。Objective: To investigate the correlation between the expression of lysophosphatidic acid(LPA) receptors and the metastasis of ovarian cancer.Methods: Invasion assay was used to detect the invasive ability of 13 ovarian cancer cell lines;Real-time RT-PCR was used to measure the mRNA expression levels of LPAR1,LPAR2 and LPAR3 in the 13 cell lines,10 samples of normal ovarian tissue,and 32 samples of epithelial ovarian cancer.Results: Seven ovarian cancer cell lines could be induced by LPA for cell invasion,which indicated as invasive cells;and the other 6 lines had poor response for LPA that indicated as non-invasive cell.All the invasive cell lines expressed high level of LPAR1,and the level was much higher than that of noninvasive cells(P=0.003).No correlation was observed between LPAR2 and LPAR3 expression and the cells' invasive ability.Among these three receptors,only LPAR2 showed higher expression level in ovarian cancer than in normal tissue(P=0.030).The expression of LPAR1 correlated with clinical stage of ovarian cancer: the higher of stages,the higher of expression levels(P=0.032);the expression of LPAR2 correlated with pathologic grade: the poorer of differentiation,the higher of expression level(P=0.020).All the cases that haD extensive abdominal metastasis showed LPAR1 expression.Conclusion: LPAR1 is the main receptor that mediates the LPA induced ovarian cancer metastasis.
