摘 要:目的探讨以开通玄府、利水解毒法组方的利开灵对脑出血后大鼠出血局部脑组织CD14、TLR4蛋白表达的影响。方法采用Rosenberg法改良后建立大鼠脑出血模型,用免疫组化方法观察48 h后不同药物干预下大鼠出血局部脑组织CD14、TLR4蛋白的表达变化,并观察利开灵对出血局部脑组织病理改变的影响。结果利开灵能有效抑制CD14和TLR4的表达,同清开灵组和安宫牛黄丸组比较,利开灵可明显降低CD14和TLR4的表达,同时可明显改善出血局部脑组织病理形态损伤。结论 CD14/TLR4信号通路可能参与了脑出血后病理损伤过程,利开灵对出血局部脑组织CD14、TLR4蛋白表达有较好的调控作用,这可能是利开灵改善脑出血后脑水肿的作用机制之一。Objective It is to explore the effect of Likailing decoction whose therapeutic method is opening sweat pore,disinhibiting water and resolving toxin on the dynamic changes of protein expression of CD14 and TLR4 in the surroundings of the bleeding region in brain edema after intracerebral hemorrhage in rats.Methods Rat models of intracerebral hemorrhage were established by improved Rosenberg method.The dynamic changes on protein expression of CD14 and TLR4 in the surroundings of the bleeding region by different drug intervention by immunohistochemistry after 48 hours were observed,and the effect of Likailing decoction on pathological morphology changes in the surroundings of the bleeding region were observed.Results Likailing decoction could effectively inhibit the protein expression of CD14 and TLR4.Compared with the Qingkailing decoction group and the Angong Niuhuang Pill group,Likailing decoction could reduce the protein expression of CD14 obviously,and had more advantages in reducing the protein expression of TLR4.Likailing decoction had the effect on lightening the injury degree on pathological morphology.Conclusion The CD14/TLR4 signal pathway may be involved in the pathological process after intracerebral hemorrhage.Likailing decoction has regulatory effect on protein expression of CD14 and TLR4 in the surroundings of the bleeding region,which may be a part of the action mechanism of improving effect of Likailing decoction on brain edema after intracerebral hemorrhage.
