摘  要:目的建立人肺癌裸鼠移植瘤模型,通过移植瘤模型可以复制不同个体的肺癌组织,用于临床和基础研究。方法人肺癌组织块接种于裸小鼠皮下,形成移植瘤后,取瘤组织做石蜡包埋切片,经HE染色,比较移植瘤来源组织与移植瘤在组织学形态上的差异。结果①成功建立人肺癌组织裸小鼠移植瘤模型,新鲜组织的成瘤率及生长速度均高于冻存组织,而成瘤潜伏时间则短于冻存组织。②移植瘤来源组织病理诊断明确,分别是鳞癌、腺癌和去分化癌各1例,移植瘤组织病理均显示为分化极差的巨细胞癌特征,没有明显的原始肿瘤特征。结论通过本研究已经成功建立肺癌裸鼠移植,并发现通过裸鼠成瘤可以使肿瘤细胞中高度恶性的细胞得到富集。Objective To establish a subcutaneous xenotransplanted tumor model of human lung cancer tissue in nude mice and in order to provide enough tissues for basic and clinical study on lung cancer. Methods Lung cancer tissues were cut into small pieces and injected subcutaneously into the nude mice, then the transplanted tumors were taken for paraffin-embedded sections and HE staining. The difference of histological manifestation between transplanted tumors and the primary lung cancer tissue was analyzed. Results Transplanted tumors of human lung cancer in nude mice were successfully established. Compared to frozen tissues, the tumor formation rate and growth velocity were higher when the fresh lung cancer tissues were injected subcutaneously into the nude mice, and its tumorigenesis time was much shorter. Although we used 3 lung cancer samples with different pathological types (1 adenocarcinoma, 1 squamous cell carcinoma, and 1 dedifferentiated carcinoma, respectively), the pathological types of all of the transplanted tumors were shown with giant cell carcinoma with poor differentiation. Conclusion Subcutaneous xenotransplanted tumor model of human iung tumor was successfully constructed in the nude mice. And highly malignant ceils can be enriched via nude mouse xenografts.
