碳酸锂对慢性应激抑郁模型大鼠海马蛋白激酶A和蛋白激酶C表达的影响  被引量:3

摘  要:目的研究碳酸锂对慢性应激抑郁模型大鼠海马蛋白激酶A(PKA)和蛋白激酶C(PKC)表达的影响。方法将大鼠随机分为抑郁模型组、碳酸锂组和对照组,每组15只。模型组和碳酸锂组给予21 d的应激刺激,此期间对照组正常饲养,刺激期间碳酸锂组每天给予碳酸锂(60 mg/kg)灌胃,模型组和对照组每天给予等体积的生理盐水灌胃。应用open-field法和液体消耗实验进行行为学检测。采用Western blot检测各组大鼠海马PKA和PKC的表达情况。结果应激后,模型组大鼠水平穿越格数、竖立次数、修饰次数和糖水消耗百分比均显著低于对照组,碳酸锂组水平穿越格数和糖水消耗百分比低于对照组(P<0.05),竖立次数和修饰次数低于对照组(P<0.01),各项数值均高于模型组,但差异无统计学意义(P>0.05)。Western blot结果显示,模型组大鼠海马PKA和PKC的表达水平显著低于对照组(P<0.01);碳酸锂组大鼠海马PKA的表达水平高于模型组(P<0.01),低于对照组(P<0.01);碳酸锂组大鼠海马PKC的表达水平高于模型组(P<0.01),略低于对照组,但差异无统计学意义(P>0.05)。结论碳酸锂可以逆转慢性应激抑郁模型大鼠海马中PKA和PKC表达的降低,碳酸锂对PKC表达的影响大于对PKA的影响。Objective To explore the effects of lithium on the expression of PKA and PKC in hippocampus of chronic stress depression rats.Methods All the rats were randomly divided into 3 groups:depression group,lithium group and control group.The rats of depression group and lithium group were given stimulation for 21 days,and meanwhile control group were bred normally.The rats of lithium group were fed with lithium(60 mg/kg),while other groups were fed with the equal volume of normal saline.Praxiology test was performed by using method of open-field and experiment of fluid consumption.The expression of PKA and PKC was detected by Western blot.Results After being stressed,the horizontal crossing numbers,the erection times,the modification times and the percentage of sacchar-consumption of the depression group rats were less than control group.The horizontal crossing numbers,the erection times,the modification times and the percentage of sacchar-consumption of lithium group were less than those of the control group(P 0.05),more than those of the depression group but no significant difference(P 0.05).In Western blot technique,the level of PKA and PKC of the depression group was less than that of the control group(P 0.01);the level of PKA of lithium group was less than that of the control group(P 0.01) and more than depression group(P 0.01);the level of PKC of lithium group was more than that of the depression group(P 0.01) and less than that of the control group but no significant difference.Conclusion Lithium can reverse the reduction of the expression of PKA and PKC in the hippocampus of chronic stress depression rats and have a stronger effect on PKC than PKA.
