摘  要:目的探讨小剂量肠血管活性肽(VIP)对急性坏死性胰腺炎(ANP)大鼠肠组织10血流量及炎性介质的影响。方法 SD大鼠96只,随机分为对照组(C组)和治疗组(T组),以5%牛磺胆酸钠于胰腺被膜下均匀注射复制ANP模型。T组经尾静脉持续滴注VIP(0.2 pmol·g^(-1)·h^(-1),持续12h),C组按同样方法注射等量生理盐水。采用放射性生物微球技术在制模后2h及12h分别测定肠组织血流量,同时检测血清TNF-α及IL-1β水平,并观察肠黏膜病理改变。结果与C组比较,T组在制模后2h及12h,肠组织血流量均明显增加(P<0.05),血清TNF-α、IL-1β水平均明显降低(P<0.05,P<0.01),肠黏膜组织损伤程度明显减轻(P<0.01)。结论在ANP早期,VIP能降低血清TNF-α及IL-1β的水平,增加肠道微循环血流量,减轻肠黏膜炎性反应。Objective To explore the protective effect of vasoactive intestinal peptide(VIP) on the changes of intestine injury,intestinal microcirculation and inflammatory mediators in acute necrotizing pancreatitis(ANP).Methods A total of 96 rats were randomized into control group(C group) and treatment group(T group).The model of ANP was established by injection of 5%sodium taurocholate into the pancreatic capsule.Little dosage of VIP(0.2 pmol ? g^(-1)?h^(-1) injected from tail veins of T group,equation dosage of NS into C group.Radioative biomicrosphere technique was applied to measure the intestinal blood flow at 2h,12h after ANP.Simultaneously,blood sample and intestine tissues were collected to observe the changes of TNK-α,IL-1βby histological exam.Results Compared with C group,the intestinal blood flow in T group increased hig her at 2h,12h after ANP(P0.05) was established.But the levels of TFN-α,IL-1βin serum were decreased(P0.05, P0.01).The intestinal pathological changes in T group were alleviated highly than that in the C group(P0.01).Conclusion In the early stage of ANP,the VIP could decrease the levels of TFN-αand IL-1βin serum,increase the intestinal blood flow,and release the inflammation in intestinal mucosa.
