摘 要:目的应用RNA干扰(RNA interference,RNAi)技术抑制VEGF的表达,观察其对B系淋巴瘤细胞株Namalwa生物学行为的影响。方法设计3条针对人VEGF mRNA的siRNA序列,经脂质体转染至Namalwa细胞。采用RT-PCR、Western blot法检测转染后Namalwa细胞VEGF mRNA和蛋白的表达。结果 siRNA-2组VEGF mRNA及其蛋白的表达明显减少(P<0.01)s,iRNA-2组细胞体外增殖能力减弱(P<0.05)。结论 RNAi技术可明显抑制Namalwa细胞VEGF的表达及细胞的体外增殖。Purpose To explore the effect of RNA interference mediated vascular endothelial growth factor (VEGF) gene silencing on biological behavior of Namalwa cells. Methods Three specific siRNAs targeting to VEGF were synthesized and transfected into B cell malignant lymphoma Namalwa cells. After dssiRNA was transfected into Namalwa cells with Lipofectamine, the proliferative ability of Namalwa cells was evaluated by MTY migration assay. The expression of VEGF mRNA and protein level was analyzed by RT-PCR and Western blot technology, respectively. Results Only siRNA-2 group have statistical significance ( P 〈 0. 01 ), it effectively inhibited the proliferation of Namalwa cells ( P 〈 0. 05 ), and decreased the expression of VEGF on both mRNA and protein level. Conclusions The siRNA targeting human VEGF gene can reduce VEGF expression, inhibit cellular proliferation and induce apoptosis in Namalwa ceils in vitro.
