NBS1基因8360G＞C(Glu185Gln)多态位点与南方人群肺癌的相关性

摘　　要：目的:研究DNA双链断裂修复基因NBSl编码区8360G>C(Glu185Gln)多态与我国南方人群肺癌发病的关联。方法:应用病例对照研究方法,采用PCR-RFLP技术检测300例肺癌患者与正常对照8360G>C多态位点的基因型,用SAS9.13软件分析该位点变异与肺癌发病的关系。结果:NBSl的8360G>C基因型频率在肺癌与对照中的分布差异有显著性(P=0.0230);携带GC变异基因型个体发生肺癌的危险性是GG基因型携带者的1.28倍(adjusted OR=1.28;95%CI=1.04-1.59;P=0.025),携带CC变异基因型个体的危险性则是1.93倍(adjusted OR=1.89;95%CI=1.43-2.51;P<0.001),且存在显著的等位基因剂量-效应关联(P_(trend)=0.0049)。结论:NBSl编码区8360C变异基因型与我国南方人群肺癌发病有关,它可能是我国南方人群肺癌发病的一个遗传相关因素。Objective:To investigate the association between the polymorphism 8360G > C in NBS1 gene(a gene for repair of double-strand DNA breaks) and the risk for lung cancer in southern Chinese population. Methods:In a hospital based case-control study,we genotyped the 8360C > C polymorphism by PCR-RFLP in 300 lung cancer cases and 300 age-and sex-matched cancer-free controls.SAS 9.13 was used to analyze the correlation between polymorphism and lung cancer risk.Results:The observed genotype frequency of 8360G > C was significantly different between the cases and controls(P =0.0230).Compared with the 8360GG genotypes carriers,the risk for development of lung cancer was 1.28-fold higher(adjusted Oft = 1.28;95%CI = 1.04 - 1.59;P=0.025) in 8360GC heterozygote carriers and 1.93-fold higher(adjusted Oft = 1.89;95%C/=1.43 -2.51;P < 0.001 ) in 8360CC homozygote carriers.There was a significant trend of dose-effect between the alleles and risk of lung cancer(P_(trend)= 0.0049).Conclusion:The polymorphism 8360G > C in NBS1 gene is linked to lung cancer and therefore may be a genetic risk factor in southern Chinese population.
