单细胞凝胶电泳法检测丹参对兔视网膜色素上皮细胞光损伤的保护作用

摘  要:目的应用单细胞凝胶电泳法(SCGE)检测丹参对家兔视网膜色素上皮细胞(RPE)可见光损伤的保护作用。方法2—4代的家兔色素上皮细胞建立光损伤模型,之后分别应用0.2mg/ml及0.8mg/ml浓度丹参液对光损伤的细胞继续培养24h,并设立无丹参培养的对照组。应用单细胞凝胶电泳法分别测定各组细胞DNA的改变。结果光损伤后的家兔RPE细胞经过丹参液培养后DNA损伤程度均有所减轻,尤其是0.8mg/ml剂量组的保护作用更为明显,细胞的尾长、彗星细胞百分率与对照组相比均有显著差异(P〈0.01)。结论丹参中的有效成分对家兔RPE光损伤有明显的保护作用,且呈现剂量依赖的量效关系。Objective SCGE detect the protection of Tan-shen on RPE cells of rabbits against photic injury. Method The 2 -4 passage RPE ceils of rabbits were adopted to establish visible light damage model, The cultured RPE cells were pretreated with taurine of 0. 2 mg/ml,0.8 mg/ml Tan-shen for 24 hours, whereas control group was pretreated without Tan-shen. The cells DNA damage of various groups were detected with single cell gel electrophoresis assay. Results The DNA damage on RPE cells of rabbits was reduced, 0.8mg/ml group have an advantage on other groups. There was great significant differece with control group in tail DNA percentage and tail length(P 〈 0.01 ). Conclusion: Tan-shen can protect RPE cells of rabbits against photic injury. The effect was does-dependent.
