牛磺酸对链脲佐菌素所致大鼠Ⅰ型糖尿病的治疗作用

摘　　要：为研究牛磺酸(Tau)对链脲佐菌素(STZ)所致糖尿病大鼠糖脂代谢及β细胞功能等方面的影响,给试验大鼠每日灌服牛磺酸,连续治疗70d,期间动态观测体重及空腹血糖。治疗第8周时处死大鼠,测定血清中C肽(C-P)、胰高血糖素(Glu)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、丙二醛(MDA)、总抗氧化能力(T-AOC)、总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白(HDL)的含量。用HE染色观察胰岛组织形态,TUNEL原位末端标记法检测胰岛细胞的凋亡百分率,用免疫组化法染色观察胰岛细胞中Fas、Bax、Bcl-2的蛋白表达水平。结果表明,各牛磺酸给药组血糖、胰高血糖素、TG、TC、MDA含量降低,体重、C肽、SOD、GSH-Px、T-AOC、HDL值升高;胰岛结构较完整,凋亡率、Bcl-2表达降低,Bax、Fas表达增高,与模型组相比,差异显著(P<0.05)。研究结果说明,牛磺酸对于糖尿病动物模型的糖脂代谢以及胰岛细胞具有一定的保护作用。To explore the effect of taurine on blood glucose,lipid metabolism,and the function of islet β cells in streptozotocin-induced diabetes mellitus rats.Diabetes mellitus rats were treated with taurine everyday,when observing their body weight and blood glucose during the 70d.The rats were killed at the 8th week of treatment,then the contents of the follow blood indexes in the diabetes mellitus rats were measured： fasting blood glucose,C-peptide（C-P）,glucagon（Glu）,superoxide dismutase（SOD）,glutathione GSH peroxidase（GSH-Px）,malondialdehyde（MDA）,total antioxidant capacity（T-AOC）,total cholesterol（TC）,triglyceride（TG）,high density lipoprotein（HDL）.Histological lesions were observed after HE stain.Apoptosis of β-cells were analyzed by way of TUNEL in situ end-labeling.At the same time,the expression of Fas,bax and bcl-2 in islet cells were observed by the way of immunohistochemistry.The results indicated that：blood glucose,glucagon levels,TG,TC and MDA of Tau group decreased,body weight,C-peptide levels,SOD,GSH-Px,T-AOC,HDL of Tau group increased;In Tau group,the structure of islet was integrity,the number of apoptosis and expression of Bcl-2 were low,the expression of Bax and Fas were high.It has significant differences（P 0.05） compared with diabetes mellitus model group.Taurine has a protective effect on blood glucose,lipid metabolism,and pancreatic islet cells.
