小鼠腹腔异位心脏移植模型的建立及注意事项

摘　　要：目的通过建立小鼠腹腔异位心脏移植模型,归纳总结制作流程和注意事项。方法选用雄性昆明鼠60只,行30对腹腔异位心脏移植模型。取供体心脏时,用4℃肝素水从下腔静脉持续灌注冲洗,待肝脏变色后,剪断腹主动脉,再持续冲洗,待流出液变淡后,进入胸腔取供体心脏,移植手术是在10倍显微镜下由单人操作完成,术中将供体小鼠心脏的升主动脉和肺动脉分别与受体小鼠的腹主动脉和下腔静脉进行端侧吻合,手术完毕后,用棉签压迫止血,1min后缓慢放开,将供体心脏置于受体腹右侧。结果共实施手术30例,成功27例,成功率为90%;移植心脏存活>30d;移植心脏5min内复跳,并且活动有力。结论通过不断优化操作流程和手术技巧,保证了小鼠心脏移植模型建立的质量及成功性。Objective While building the heterotopic heart transplant models in mice,we summarizes skill details of operation and notice.Methods The donors and receptors were all kuming mouse,we are perfusing and washing continuously the heart of donors,ascending aorta and pulmonary artery of donor Respectively were anastomosed with Abdominal Aorta and inferior vena cava of receptor by end-side ansatomosis during operation.Results We significantly improved the successful and survival rate,and it could reach to 90%.Conclusion By condensed routine daily practice of surgery and summarization of skill details,ensure the quality of the model and survival rate in recipients.
