食管鳞癌组织中磷脂酶A_2 mRNA检测与分析

摘　　要：目的探讨分泌型磷脂酶A2-Ⅱa(secretory phospholipase A2-Ⅱa,sPLA2-Ⅱa)及胞浆型磷脂酶A2(cytosolic phospholipase A2,cPLA2)mRNA表达与食管癌发生、发展及浸润、转移的关系。方法应用原位杂交法检测45例食管鳞癌组织、22例癌旁不典型增生组织及45例正常食管黏膜组织中sPLA2-Ⅱa及cPLA2 mRNA表达。结果食管鳞癌组织中sPLA2-Ⅱa mRNA表达与癌的浸润深度、淋巴结转移及TNM分期密切相关(P<0.05);在食管鳞癌癌变过程中sPLA2-Ⅱa mRNA表达在正常黏膜组织、癌旁不典型增生组织及癌组织中的表达率依次增高,组间比较差异有统计学意义(P<0.05)。cPLA2 mRNA表达与癌的浸润深度、淋巴结转移及TNM分期均密切相关(P<0.05);cPLA2 mRNA在正常食管黏膜组织、癌旁不典型增生组织及癌组织中的表达率依次增高,组间比较差异有统计学意义(P<0.05),sPLA2-Ⅱa及cPLA2的表达呈正相关(rp=0.429,P=0.003)。结论 sPLA2-Ⅱa及cPLA2 mRNA在食管鳞癌组织中表达显著升高,并与食管鳞癌生物学行为关系密切,提示sPLA2-Ⅱa及cPLA2低表达与食管鳞癌的发生、发展有关,sPLA2-Ⅱa及cPLA2可作为食管鳞癌早期诊断和判断预后的辅助指标。Objective To investigate the correlations between the mRNA expression of sPLA2-Ⅱa and cPLA2 protein and the occurrence,development,invasion and metastasis of esophageal squamous carcinoma.Methods In situ hybridization method was applied to detect the mRNA expression of sPLA2-Ⅱa and cPLA2 in 45 samples of esophageal squamous carcinoma,22 samples of adjacent atypical hyperplasia epithelium and 45 samples of normal esophageal epithelium.Results The expression of sPLA2-Ⅱa mRNA was significantly correlated with the infiltration,lymphatic metastasis and TNM grade of esophageal squamous carcinoma（P0.05）.The postive rates of sPLA2-Ⅱa mRNA in normal esophageal epithelium,adjacent atypical hyperplasia epithelium and carcinoma were significantly different among groups（P0.01）.The expression of cPLA2 mRNA was significnatly correlated with the infiltration,lymphatic metastasis and TNM grade of esophageal squamous carcinoma（P0.05）.The positive rates of cPLA2 mRNA in normal esophageal epithelium,adjacent atypical hyperplasia epithelium and carcinoma were significantly different among groups（P0.01）.There was significantly positive correlation between the expression of sPLA2-Ⅱa and cPLA2（rp=0.429,P=0.003）.Conclusion sPLA2-Ⅱa and cPLA2 play important roles in the infiltration,metastasis and oncogenesis of the mucosa epithelium in esophageal carcinoma.Combined examination of sPLA2-Ⅱa and cPLA2 could be applied in early diagnosis and prognosis assessment of esophageal squamous carcinoma.
