摘 要:目的 研究糖化血清蛋白对培养的肾小球系膜细胞生长和分泌 型胶原的影响 ,以了解糖化蛋白与糖尿病肾病的关系。方法 正常人血清在 2 8mmol L葡萄糖的磷酸缓冲液中 2 5℃孵育 7天 ,使血清蛋白糖化 ,分别测定不同浓度的糖化血清蛋白对肾小球系膜细胞 (MC) 3 H Thymidine掺入和培养上清液中 型胶原含量的影响。结果 糖化血清蛋白浓度为 3 %和 10 % ,可明显抑制3 H Thymidine掺入量且呈剂量依赖性。与相同剂量的正常血清比 ,分别抑制 19%和66% ,而 型胶原分泌量分别增加 2 4 %和 3 5 6%。结论 糖化血清蛋白可能通过减少肾小球系膜细胞形成和增加系膜基质扩张而促进糖尿病肾脏的肾小球硬化。Objective To study the relationship between glycated serum proteins and development of diabetic nephropathy by investigating effects of glycated serum proteins on mesangial cells.Methods Normal human sera was nonenzymatically glycated by incubation for 7 days with 28mmol/L glucose. The amount of 3H Thymidine incorporation into mesangial cells and secretion of type Ⅳ collagen in media were respectively measured when rat mesangial cells had been cultured with different concentrations of glycated serum proteins for 48 hours.Results Supplementation of mesangial cells cultured in the presence of 3% and 10% glycated serum protein inhibited 3H Thymidine incorporation by 19% and 66% respectively comparing with the same concentrations of normal serum, which was dose dependent on glycated serum proteins, and increased content of type Ⅳ collagen by 24% and 35 6% respectively.Conclusion The glycated serum proteins played an important role in development of diabetic nephropathy by causing the expansion of extracellular matrix in mesangial membrane area and inducing glomerulosclerosis.
