摘 要:目的建立羌活饮片挥发油的气相色谱-质谱(GC-MS)数字化指纹图谱,以控制羌活饮片的质量。方法用水蒸气蒸馏法提取样品的挥发油后,采用GC-MS联用法测定挥发油的化学成分。色谱条件为采用HP-5毛细管色谱柱(30 m×0.25 mm,0.1μm);载气为氦气;体积流量1.0 mL/min;分流比50∶1;进样温度270℃。程序升温方式为初始柱温70℃,维持3 min,以10℃/min升至250℃,维持5 min。结果建立了11批羌活饮片挥发油化学成分的GC-MS数字化指纹图谱,标定了40个共有峰,占总色谱峰面积的80.8%~90.6%。其中峰面积较大的化合物为1R-α-蒎烯,β-蒎烯,桉油醇,水菖蒲酮,D-柠檬烯,4-松油醇,对伞花烃,乙酸龙脑酯,α-松油醇,γ-松油烯,龙脑,布藜醇。结论所建立的GC-MS指纹图谱能用于羌活和宽叶羌活饮片的鉴定和质量控制。且方法灵敏度高,重复性良好。AIM To establish the digitized fingerprint of the essential oils of processed Notopterygii Rhizoma et Radix by GC-MS for the quality control. METHODS The essential oil were obtained by steam distillation, and chemical constituents and fingerprint of the essential oils were determined by GC-MS. The chromatography analysis was carried out on an HP-5 capillary column (30 m×0.25 mm,0.1μm) , by using helium as the carries gas, the flow rate of 1.0 mL/min, the split ratio 50 : 1, sample inlet temperature of 270℃. The temperature program was as follows: 70℃ for three minutes, rose to 250℃ at 10℃/min, then retained at 250℃ for five minutes. RE- SULTS The GC-MS digitized fingerprint of the essential oils from 11 batches of processed Notopterygii incisi Rhi- zoma et Radix contained 40 common peaks, and was composed of 80.8% -90.6% of the total peak area. Some of these major compounds consisted of 1R-αpinene, β-pinene, eudesmol, shyobunone, D-limonene, 4-terpineol, p- cymene, bomyl acetate, α-terpineol, γ-terpinene, borneol, and butnesol. CONCLUSION The fingerprint can be used for the identification and quality control of processed Notopterygii incisi Rhizoma et Radix and Notopterygii forbesii Rhizoma et Radix, and it also has high sensitivity and good reproducibility.
