摘 要:目的 探讨20 个植物药对肿瘤细胞DNA多聚酶的作用及对肿瘤细胞生长的影响。方法 自对数生长期的小鼠艾氏腹水癌细胞中提取核酶液,根据Ono 法,以3H-dTTP掺入活化小牛胸腺DNA的量作为DNA 多聚酶活性指标。用MTT法测定药物对肿瘤细胞生长的影响。结果 巴马汀和小蘖碱抑制小鼠艾氏腹水癌细胞DNA 多聚酶,在100 μg/m l浓度下,巴马汀抑酶率为68.5 % ±6.0 % ,小蘖碱为68.6 % ±15.3 % ,其余18 个植物药均不抑制DNA多聚酶。小蘖碱对体外培养的人早幼粒细胞白血病HL60 细胞和人类淋巴母细胞CCRF-CEM 细胞均有抑制作用,IC50 分别为5.14 μg/m l和34.55 μg/m l。结论 巴马汀和小蘖碱均是肿瘤细胞DNA多聚酶的抑制剂。小蘖碱对体外培养的肿瘤细胞有抑制作用。对DNA 多聚酶的抑制作用可能是其抗瘤机制之一。Objective To investigate the effect of 20 plant drugs on DNA polymerase activity and cell growth of tumor.Methods Crude nuclear enzyme was extracted from logarithmic growing Ehrlich ascites tumor cells according to Ono′s method.Quantity of 3H dTTP incorporating into calf thymus DNA was used to detect the activity of DNA polymerase.MTT method was used to test the effect of drugs on growth of tumor cells.Results Palmatine and Berberine inhibited DNA polymerase of Ehrlich ascites cancer cells,the inhibition rate was respectively as 68.5 %±6 0 % and 68.6 %±15 3 % under the concentration of 100 μg/ml. Other 18 drugs did not inhibit DNA polymerase.Berberine inhibited tumor cell growth in vitro in HL 60 cells and CCRF CEM cells,the IC 50 value was respectively as 5.14 μg/ml and 34.55 μg/ml.Conclusion Palmatine and Berberine are inhibitors of DNA polymerase of tumor cells.Berberine inhibits tumor cells in vitro.The inhibition on DNA polymerase may be one of the mechanisms of antitumor action by Berberine.The screening method with DNA polymerase as a target may be a supplement of present screening methods for antitumor drugs.$$$$
