摘  要:目的 体外研究维甲类化合物的衍生物ATRA对LoVo 结肠癌细胞株的作用。方法 观察ATRA作用下LoVo 细胞的增殖、细胞周期和凋亡。结果 ATRA使细胞增殖抑制,细胞阻滞于G0 /G1 期,高浓度组出现亚“G0/G1 ”峰。ALP比值增高,高浓度组电镜下见凋亡细胞,TDT法测凋亡细胞比例增高,但传6 代后不增加。结论 提示ATRA可诱导LoVo 细胞分化及凋亡,但有一定限度;实验结果为临床应用ATRAObjective The potential roles of all trans retinoic acid (ATRA) in colorectal cancer cell line LoVo in vitro were studied.Methods The proliferation, the cell cycle and the apoptosis of LoVo cells were determined after treated with ATRA.Results ATRA could significantly inhibit proliferation of LoVo and greatly increase ALP of the cells in time dependent manner.In cytometry analysis,LoVo cells lines accumulated in G 1 phase and appeared an extra “sub G 0/G 1 peak” when higher dosage of ATRA was presented.Apoptotic cells only in 10 -5 mol ATRA were found by electronic microscopy.Conclusion ATRA could both induce differentiation and apoptosis of LoVo cells.This observation may become an indicator for clinical trials on prevention of colorectal cancer.$$$$
