摘  要:[目的]研究从淋浆水中提取SOD酶的工艺条件。[方法]以陕西太白酒厂酿酒后的淋浆水为材料,比较甲苯、异丙醇和乙醇-氯仿3种有机溶剂从淋浆水中提取SOD粗酶液的效果,研究温度和pH对SOD酶活力的影响以及不同异丙醇浓度对SOD提取的影响,采用丙酮2次沉淀对SOD进行分离纯化,并采用H2O2和乙醇-氯仿对样品SOD酶类型进行敏感性鉴定。[结果]试验确定以80%的异丙醇为最佳粗酶液提取条件。丙酮2次沉淀得到纯化的SOD产品,其酶比活为1 026 U/mg,酶回收率达65.2%。该酶经H2O2和乙醇-氯仿敏感性鉴定为Cu.Zn-SOD,对热和酸碱度具有一定稳定性,在50℃以下,酶相对活性保持在91.2%~100%,pH为3.0~10.0时SOD活性保持在74.1%~93.6%。[结论]该提取工艺过程简便,易操作,是较为理想的从淋浆水中提取SOD的工艺。[ Objective] The objective was to study the technology conditions for extracting SOD from the yellow seriflux. [ Method J With the yellow seriflux after brewing the wine in Shaanxi Taibai White Wine Plant as the materials, 3 kinds of organic solvents including the methylbenzene, isopropanol, ethanol-chloroform were compared on the effects of extracting the SOD crud enzyme liquid from the yellow seriflux, the influences of the temperature and pH value on the SOD enzyme activity and the influences of isopropanol with different concen, on the SOD extraction were studied, the SOD was made for the separation and purification by 2 times of precipitation with the acetone, and the type of sample SOD enzyme was conducted for the sensitive identification by suing the H202 and ethanol-chloroform. [ Result] Through test it was confirmed that the 80% isopropanol was the optimum condition for extracting the crud enzyme liquid. The purified SOD product was obtained by 2 times of precipitation with the acetone, with the specific activity of 1 026 U/mg and the recovery rate of 65.2%. The enzyme was identified as Cu ·Zn-SOD through the sensitivity tests with HE 02 and ethanol-chloroform and showed some stability to heat and pH, meanwhile, when the temperature was under 50 ℃, the relative SOD activity could keep constant at 91.2% - 100% and when the pH was 3.0 - 10.0, the SOD ac- tivity could keep in 74.1% -93.6%. [ Conclusion ] The extraction process was simple and easy to operate, which was the ideal process of extracting SOD from the yellow seriflux.
