摘　　要：目的探讨STAT3对肺腺癌早期诊断的价值及能否成为一种新的肺肿瘤标志物。方法采用荧光定量PCR、免疫组织化学PV法分别检测人肺腺癌细胞A549和正常人胚肺细胞MRC-5中STAT3、CEA、CA125 mRNA和蛋白表达情况,分析在人肺腺癌细胞A549中STAT3 mRNA分别与CEA、CA125 mRNA表达的相关性。结果(1)STAT3、CEA、CA125 mRNA在人肺腺癌细胞A549中的表达均明显高于它们各自在正常人胚肺细胞MRC-5中的表达,且STAT3 mRNA的表达与临床上常用的肺肿瘤标志物CEA、CA125 mRNA的表达均呈正相关。(2)STAT3在人肺腺癌细胞A549诊断中的敏感性(92.2%)与准确性(91.3%)均高于CEA、CA125。结论 STAT3有望成为一种新的肺肿瘤标志物用于临床。Objective To explore the value of STAT3 on early diagnosis of lung cancer and the possibility as a new tumor marker. Methods The expressions of protein and mRNA of STAT3,CEA and CA125 in lung adenocarcinoma cell A549 and human normal lung cell MRC-5 were tested by real-time PCR and immunohistochemistry staining（PV）.The correlation between STAT3mRNA and CEAmRNA,CA125mRNA in lung adenocarcinoma cell A549 was analyzed. Results（1） The mRNA levels of STAT3,CEA and CA125 in lung adenocarcinoma cell A549 were apparently higher than those in human normal lung cell MRC-5.The mRNA level of STAT3 was in a positive correlation with CEA and CA125 in lung adenocarcinoma cell A549.（2） STAT3 showed the highest sensitivity（92.2%） and accuracy（91.3%） for the diagnosis of lung adenocarcinoma cell A549. Conclusions STAT3 can be served as a new marker of lung cancer and be recommended for clinical use.
